This report contains a study to investigate the acute toxicity of BENZANIL RUBINE 2BL (Direct Red 83-1) to rainbow trout (oncorhynchus mykiss). The objective of this study was to determine the 96 hr median lethal concentration (LC50). 5 concentrations were tested 10, 18, 32, 56 and 100 mg/l.

The results of this study indicate that at concentration above 18 mg/l is fairly rapidly lethal to rainbow trout. The estimated median period of survival, of the exposed fish, obtained for at levels of 56, 32 and 18 mg/l, were 39hr, 57hr and 47hr respectively. The lowest test concentration which caused 100% mortality of exposure fish within 96 was 18 mg/l. The highest no-observed lethal effect concentration of the test substance to rainbow trout could not be determined as there was 42.9% mortality at the lowest test concentration.

Dilution of the sample from 18 mg/l to 10 mg/l significantly increases the survival time of the trout. The estimated LC50 values of 59 mg/l at 24 hrs and 13 mg/l at 96 hrs support these observations.

A mortality of 0% was observed in the control tank at the end of the test period. This is within the range of mortality acceptable for validation of the test results.

The temperature of the control and test tanks was slightly above the recommended limit towards the end of the test, but as there was no pattern of increase in temperature, this was not thought to have adversely affected the data.
