A concentrated stock solution of the test substance was be prepared as appropriate in defined Elendt M4 dilution medium. This stock solution was be diluted to achieve the required nominal test substance concentrations.

The Daphnia magna (Straus) strain held at Huntingdon Life Sciences was supplied by Sheffield University, having been originally obtained from the National Institute for Applied Chemical Research (IRCHA), France. Daphnia cultures were established with juvenile Daphnia taken from existing cultures after the first brood had been removed and discarded. Cultures are maintained in defined Elendt M4 dilution medium (between 500 and 1500 mL) in glass beakers with glass or perspex covers. The cultures were maintained in a temperature controlled laboratory at 20 +/- 2 degrees C. The day-length was controlled giving a photoperiod of 16 hours light, provided by overhead fluorescent tubes, and 8 hours dark. The prepared test substance dilutions in defined medium were be placed into the test vessels. The test vessels were not aerated during the test. The vessels were of sufficient size to meet the minimum guideline criterion of one Daphnia per 2 mL of test substance dilution. Twenty Daphnia, less than 24 hours old, were used at each test substance concentration in the definitive test. Five Daphnia were placed in each vessel. The Daphnia were not fed during the exposure. Observations were made after 24 and 48 hours of exposure and the numbers of floating, mobile and immobile Daphnia were recorded. For the test to be valid, the number of floating and/or immobile Daphnia in the control group must not exceed 10% at the end of the test.

Significant complete immobilization (20/20) was observed at a nominal Crude 1,3-dichloropropene concentration of 10 mg/L at 24 hr of exposure. Complete immobilization of the Daphnids was observed at 4.55 and 10 mg/L following 48 hr of exposure. Immobilization was 1/20 at 2.1 mg/L and 6/20 at 4.55 mg/L at 24 hr. The immobilization was 1/20 at 0.94 and 2.1 mg/L after 48 hr of exposure.
