Appearance/physical state/colour:

On the basis of physical observation, Dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium, disodium salt was found to be a dark green colored powder.

Melting point/freezing point:

The melting point determination of dihydrogen(ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium,disodium salt (CAS No. 2353-45-9) was done following OECD Guideline 102 and using heating metal block apparatus. The test was conducted in replicates and mean melting point range was found to be 265°C to 275°C at 977.1 hPa.With melting, the color of test item changed from dark green to orange.

Boiling point:

The boiling point determination of dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium, disodium salt (CAS No. 2353-45-9)was done following OECD Guideline 103 and according to the method of Siwoloboff. A marked color change was observed at 258 C when the sample started melting from dark green to orange and formed a paste like appearance. The test was conducted in replicates and mean boiling point was found to be greater than 260 °C at 977.3hPa.

Density:

The density of test item dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium,disodium salt (CAS No. 2353-45-9)was determined following OECD Guideline 109 and accordingto themethod recommended in the International Pharmacopoeia, fourth Edition, 2014 using bulk density apparatus and 250 ml measuring cylinder. The pour and tapped density of the test item was found to be 0.5131 g/cm3and 0.6250 g/cm3at 27°C and 978.8 hPa.

Particle size distribution (Granulometry):

The particle size distribution of test item Dihydrogen(ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium,disodium salt (CAS No.2353-45-9) was determined by granulometric analysis with rotap sieve shaker. The particle size distribution was found to be in the range of 150 micron to 25 micron.

The water solubility determination of test item dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene] cyclohexa-2,5-dien-1-ylidene] (3-sulphonatobenzyl)ammonium, disodium salt(CAS No. 2353-45-9) was done gravimetrically as per the shake flask method using Whatman filter paper No.1. The solubility was found to be 69.30 g/L at 24oC.

Surface tension

In accordance with column 2 of Annex VII of the REACH regulation, this end point was not considered for testing since surface activity is not a desired property for the chemical (FD&C Green 3) which finds use as a food color, tissue and protein stain and cosmetic coloring agent.

Flash point:

The flash point determination of Test item dihydrogen(ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium,disodium salt (CAS No. 2353-45-9) was done. The method is closed cup method using Pensky-Martens Flash Point Apparatus. The flashpoint of the test item was found to be 178°C at 977.05hPa

Auto flammability

The chemical dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium, disodium salt (Synonym: Fast Green FCF) is used as a tissue and protein stain, besides having use as a colorant for active pharmaceutical ingredients and food applications and has a flash point > 100oC. Thus, considering the high flash point, the test chemical is not expected to exhibit auto flammability.

Flammability

The chemical dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium, disodium saltdoes not indicate any ignition when the flame of bunsen burner having temperature of approximately 950°C is brought in contact with it. Thus it can be concluded that benzimidazole is not flammable at high temperature. However since such high temperatures are not found under normal circumstances of transportation and use, thus benzimidazole was considered non-flammable for chemical safety assessment.

Explosiveness

This end point was considered for waiver since in accordance with column 2 of Annex VII of the REACH regulation, there are no chemical groups associated with explosive properties present in the molecule of Dihydrogen (ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium, disodium salt (Synonym FD&C Green 3, Fast Green FCF)

Oxidising properties

In accordance with column 2 of Annex VII of the REACH regulation, this study was considered to be scientifically unjustified since the chemical FD&C Green 3/Fast Green FCF is incapable of reacting exothermically with combustible materials based upon its chemical structure. The chemical is organic, contains oxygen but no halogen atoms and the oxygen atoms are not chemically bonded to nitrogen or oxygen. Thus the chemical is not expected to exhibit oxidising properties.

pH:

The pH determination of test item Dihydrogen(ethyl)[4-[4-[ethyl(3-sulphonatobenzyl)amino](4-hydroxy-2-sulphonatobenzhydrylidene]cyclohexa-2,5-dien-1-ylidene](3-sulphonatobenzyl)ammonium,disodium salt (CAS No.2353-45-9) was done following OECD guideline 122. pH of 1% w/v aqueous solution was measured with the help of pH probe and it was observed to be 4.0 at 29.8°C.
