Description of key information
The oral LD50 of 2-vinylpyridine in rats is 336 mg/kg body weight (bw), and when 2-VP is applied dermally to rabbits under occlusive dressings, the LD50 value is 640 mg/kg bw. As there are no accepted methods of extrapolating from lethal values in animals to "no effect" levels in humans, the risk assessment will be qualitative. Risk management measures in place for skin and eye corrosion will protect against acute dermal toxicity in humans.

Additional information
The LD50 of 2-vinylpyridine in rats is 336 mg/kg body weight (bw), when dissolved in water. The LD50 is higher when the substance is suspended in corn oil and administered by oral gavage to rats. When 2-VP is applied dermally to rabbits under occlusive dressings according to EPA guideline protocols, the resulting LD50 value is 640 mg/kg bw. Due to its corrosive nature, 2VP was not studied in inhalation toxicity protocols in vivo.
Justification for classification or non-classification
2-Vinylpyridine is classified, according to Regulation (EC) No. 1272/2008, as Acute Toxicity Category 4 for oral toxicity, as the LD50 of 336 mg/kg bw in the rat falls within the range of 300 to 2000 mg/kg bw. For acute dermal toxicity, 2-Vinylpyridine is classified as Acute Category 3, as the LD50 value of 640 mg/kg bw in the rabbit falls between 200 and 1000 mg/kg bw. Due to its corrosive nature, it is also classified as H071, Corrosive to the respiratory tract. According to Directive 67/548/EEC, 2-vinylpyridine is classified as Xn, R21/22, (Harmful).
