Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Genetic toxicity in vitro

Description of key information

According to the umu test with or without metabolic activation (S9 mix), the test item Gallium trichloride is not mutagenic.

Reference
Endpoint:
in vitro gene mutation study in bacteria
Type of information:
read-across from supporting substance (structural analogue or surrogate)
Adequacy of study:
key study
Reliability:
1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
guideline study
Justification for type of information:
As gallium is quite insoluble (ca 46 µg/L) toxicity testing has been performed with gallium(III) chloride, which has a higher water solubilty. A read-across approach is followed where the effects of gallium (III) chloride hydrate are completely attributed to the gallium ion. Because both AsCl3 and As release As ions upon dissolution and the higher solubility of AsCl3, this is a conservative, worst-case approach for gallium.
Reason / purpose for cross-reference:
read-across source
Qualifier:
according to guideline
Guideline:
JAPAN: Guidelines for Screening Mutagenicity Testing Of Chemicals
Version / remarks:
umu test
GLP compliance:
not specified
Type of assay:
SOS/umu assay
Species / strain / cell type:
S. typhimurium TA 1535
Metabolic activation:
with and without
Metabolic activation system:
S-9 mix: a mixture of the microsome fraction of a rat liver with coenzymes such as reduced nicotinamide adenine dinucleotide phosphate and reduced nicotinamide adenine dinucleotide, was used
Untreated negative controls:
yes
Remarks:
PBS
Positive controls:
yes
Positive control substance:
furylfuramide
other: 2-aminoanthracene
Key result
Species / strain:
S. typhimurium TA 1535
Metabolic activation:
with and without
Genotoxicity:
negative
Cytotoxicity / choice of top concentrations:
no cytotoxicity
Conclusions:
The test item Gallium trichloride is not mutagenic, with or without metabolic activation (S9 mix). Because both AsCl3 and As release As ions upon dissolution and the higher solubility of AsCl3, this results is considered also applicable for gallium metal.
Executive summary:

The test item Gallium trichloride is not mutagenic, with or without metabolic activation (S9 mix). Because both AsCl3 and As release As ions upon dissolution and the higher solubility of AsCl3, this results is considered also applicable for gallium metal.

Endpoint conclusion
Endpoint conclusion:

Justification for classification or non-classification

According to the umu test with or without metabolic activation (S9 mix), gallium is not mutagenic.
