- Concentrations: 100, 180, 320, 560, and 1000 mg/kg
- Sampling method: At the start of the test, 10 earthworms were placed in each test and control vessel on the artificial soil, at random. The test vessels were covered with plastic film with ventilation holes and maintained in a temperature controlled room at 21°C under continuous illumination at 548 to 720 lux. The earthworms were not individually identified and received no food during exposure. Any mortalities and sub-lethal effects of exposure were recorded 7, and 14 days after the start of treatment. The criterion of death was taken to be the absence of any reaction to a physical stimulus at either end of the body. Any unaccounted for worms were classed as mortalities.

- Method of mixing into soil (if used): Amount of test material were each separately incorporated into 2.5 kg (dry weight) of artificial soil with 650 mL of deionised reverse osmosis water and mixed using a Hobart QE 200 mixer to give the test concentrations with a nominal moisture content of 30% of dry weight. The control was prepared in an identical manner without the addition of test material.
- Controls: An amount of soil was taken for moisture content determination from the control and each test group. The remaining soil was dispersed to four replicates R1-R4.

Executive summary:

A study, according to OECD Guideline 207 and EEC Commission Directive 87/302/EEC, was performed to assess the acute toxicity of the test material to the earthworm (Eisenia foetida) in an artificial soil.

Following a preliminary range-finding test (test concentrations of 10, 100 and 1000 mg/Kg), earthworms were exposed in groups of 40 (4 replicates of 10 worms per concentration) to a range of concentrations of 100, 180, 320, 560 and 1000 mg/Kg of soil for a period of 14 days. The number of mortalities was determined after 7 and 14 days exposure.

A positive control study using chloroacetamide, conducted approximately every 6 months, was reported for reference purposes.

The 14d-LC50 for the test material to earthworms based on nominal test concentrations was 250 mg/Kg (95% confidence limits = 240 -260 mg/Kg).

The result of the positive control study gave a 14d-LC50 for chloroacetamide of 23 mg/Kg (95% confidence limits = 20 -25 mg/Kg).
