Materials and methods

Principles of method if other than guideline:
A DCB (DNA-cell-binding) assay was performed. The DCB investigated the formation of complexes of DNA with proteins and with other nucleic acid molecules in the presence of mutagens. The formation of adducts was detectable by the decreased yields of DNA recovered into water phase during cold phenol extraction of the cellular lysate of Ehrlich ascites cells.

If more than 1 % of DNA was found attached to intact animals cells (after subtracting the control values), such level of binding was considered indicative of a positive response. If the levels of binding in the chemical-containing tube were below those of the control sample, the figure "0.0" was given as result. For the test substance, less than 1% of DNA was found attached indicating a negative result.
