HPLC CONDITIONS: Chemicals injected onto a cyano column move along it by partitioning between the mobile phase and the stationary phase.
Column: Apex cyano, 5µ, 25 cm x 4.6 mm id.
Mobile phase: Methanol/water, 40/60 v/v.
Flow rate: 1.0 ml/min.
Pressure: 3500 psi.
Analytical wavelength: 210 nm.
Injection volume: 50 µl
Chart speed: 5 mm/min.
Range: -50 to 200 mV.
Column temperature: 20 °C +/- 0.5 °C

DETERMINATION OF DEAD TIME
The dead time (the average time a solvent molecule needs to pass through the column) was taken as 2.469 minutes, the time of the earliest consistently appearing peak (mean of 2.467 and 2.470). Thiourea (a recommended organic compound for the measurement of the dead time) was used and was found to be slightly retained on the column.

REFERENCE SUBSTANCES
- Identity: fenthion, monolinuron, sulprofos, triadimenol, triazophos and trifluralin.

DETERMINATION OF RETENTION TIMES
- Quantity of test substance introduced in the column: test A 47.3 µg, test B 47.9 µg

EVALUATION
- Calculation of capacity factors k': k= t-to/to
- Determination of the log Koc value: log Koc= logk-I/S
