Test material

Constituent 1
Chemical structure
Reference substance name:
2-butoxyethanol
EC Number:
203-905-0
EC Name:
2-butoxyethanol
Cas Number:
111-76-2
Molecular formula:
C6H14O2
IUPAC Name:
2-butoxyethanol
Details on test material:
An industrial cleaner. Analysis of the mixture showed the presence of 2-butoxyethanol, trisodium phosphate, nonylphenol polyethylene glycol ether, anthraquinone, lemon fragrance and an antifoam agent.

Method

Type of population:
occupational
Subjects:
CASE 1
- Number of subjects exposed: 1
- Sex: female
- Age: 49
- Race: white

CASE 2
- Number of subjects exposed: 1
- Sex: male
- Age: 43
- Race: white
Clinical history:
CASE 1: History of erythematous, papular eruption on hands, face and trunk. Treated with corticosteroids successfully. As this was resolving, intense pruritic vesicular eruptions developed on elbows, knees, abdomen. Biopsy findings found to be consistent with DH.
CASE 2: several hours after exposure, intense burning and tingling in hands, puritic eruption of grouped vesicles on hands and forearms. Biopsy findings found to be consistent with DH.
Controls:
no data
Route of administration:
dermal
Details on study design:
Patch testing on unaffected skin of the back was carried out with the individual chemicals of the mixture and at the same concentration than in the mixture (except for 2-butoxyethanol which was diluted to avoid irritation)

Results and discussion

Results of examinations:
There was no reported reaction to 2 -butoxyethanol.

Applicant's summary and conclusion

Conclusions:
2-butoxyethanol was not the cause of dermatitis herpetiformis in two occupational case studies.
Executive summary:

Two cases of dermatitis herpetiformis (DH) temporally related to the use of a cleaning solution containing 2 -butoxyethanol (2BE) have been reported. Analysis of the mixture showed the presence of 2 -butoxyethanol amongst a number of other materials. Patch testing was carried out with the individual chemicals at the same concentration than in the mixture, except for 2BE which was diluted to avoid irritation). There was no reported reaction to 2 -butoxyethanol in the patch tests whereas some of the other chemicals caused an adverse response.
