The physico-chemical and biological reactions of different concentrations of DEMB were tested on non-living animal-skin as a pre-test that was performed for the guinea pig maximization test. Removed dorsal skin of 3 rats was treated with 10, 200 mg/mL and undiluted test substance. No biological reactions could be observed in rat skin.

Additionally, guinea pig skin was treated in situ with 10, 200 mg/kg and undiluted DEMB by intradermal injection. Moreover, 200 mg/mL and undiluted DEMB were applied occlusively to a clipped flank. Two hours later, the dressing was removed and skin was analyzed. Treatment with DEMB resulted in skin corrosion, intradermal injections showed a dose-dependent increase of the diameter of necrotic tissue.

Justification for classification or non-classification

According to Annex VI of the EU Regulation (EC) N0. 1272/2008 (CLP) and Annex I of the EU directive 67/548/EEC, DEMB is classified as skin corrosive Cat 1B and R34, respectively.
