Short term toxicity to fish

In a three weight of evidence studies:

ECOSAR Version 1.11:

For freshwater fish:Based on the QSAR prediction done using the EPI Suite ECOSAR version 1.11, the 96 hours LC50 was estimated to be 0.18 mg/L on fish for substance Benzenamine, N-phenyl-, styrenatedwith mortality effects. Thus based on this value it can be concluded that the substance can be classified asaquatic acute category 1as per the criteria of CLP regulation.

For saltwater fish:Based on the QSAR prediction done using the EPI Suite ECOSAR version 1.11, the 96 hours LC50 was estimated to be 0.231 mg/L on fish for substanceBenzenamine, N-phenyl-, styrenatedwith mortality effects. Thus based on this value it can be concluded that the substance can be classified asaquatic acute category 1as per the criteria of CLP regulation.

U. S. EPA - High Production Volume (HPV) Challenge Program; September, 2009:The short term toxicity of Benzenamine, N-phenyl-, and styrenated was examined on species Brachydanio rerio. The benzenamine, N-phenyl-, styrenated was dispersed in water by means of an Ultra-Turrax. Mortality increased with increasing concentration of benzenamine, N-phenyl-, styrenated added to the test solution and the sponsor reported 96-h LC50 of 920 mg/L for Benzenamine, N-phenyl-, styrenated in Brachydanio rerio under static condition.

 

But the substance is not readily biodegradable in water also BCF ≥ 500 and log Kow ≥ 4 which not support to non toxic condition.Thus target chemical consider for the classification in aquatic toxicity as per the CLP criteria.

Thus overall all studies indicate that the test substance Benzenamine, N-phenyl-, styrenated can be classified as aquatic acute category 1 for fish as per the criteria of CLP regulation.

Short term toxicity to aquatic invertebrates

Various studies for the target substanceBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2)along with its predicted data were reviewed to summarize the following information:

48 hrs aquatic toxicity study (SSS QSAR prediction model, 2016) was conducted to assess toxic effects of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) and the results were predicted. The study was based on the effects of the test compound on the Daphnia magna (life stage – larvae) in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compoundBenzenamine, N-phenyl-, styrenatedwas estimated to be 0.0507 mg/L on the basis of intoxication.

48 hrs aquatic toxicity study (EPI suite, 2016) was conducted to assess toxic effects of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) and the results were predicted. The study was based on the effects of the test compound on Daphnia magna in a static fresh water system. The predicted data suggests the LC50 for the test compoundBenzenamine, N-phenyl-, styrenatedwas estimated to be 0.14 mg/L on the basis of mobility. Thus based on this value it can be concluded that the substance can be classified as Aquatic acute category 1 as per the criteria of CLP regulation.

Based on theoverall reported resultsof various studies of short term toxicity to aquatic invertebrates for target substance, it can be concluded thatthe substance can be classified as aquatic acute category 1 as per the criteria of CLP regulation.

 

Toxicity to aquatic algae and cyanobacteria

Various studies for the target substance Benzenamine, N-phenyl-, styrenated (CAS No. 68442-68-2) along with its predicted data were reviewed to summarize the following information:

96 hrs aquatic toxicity study (EPI suite, 2016) was predicted to assess toxic effects of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2)and the results were predicted. The study was based on the effects of the test compound on green algae in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2)was estimated to be 0.39 mg/L on the basis of growth rate.Thus based on this value it can be concluded that the substance can be classified as Aquatic acute category 1 as per the criteria of CLP regulation.

 

72 hrs aquatic toxicity study (SSS QSAR prediction model, 2016) was predicted to assess toxic effects of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) and the results were predicted. The study was based on the effects of the test compound on Scenedesmus subspicatus in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) was estimated to be 1.5639 mg/L on the basis of growth rate. Thus based on this value it can be concluded that the substance can be classified as Aquatic chronic category 2 as per the criteria of CLP regulation.

72 hrs aquatic toxicity study (Danish (Q)SAR Database, 2016) was predicted to assess toxic effects of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) and the results were predicted. The study was based on the effects of the test compound on Pseudokirchneriella spp. in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2) was estimated to be 2.369 mg/L on the basis of growth rate. Thus based on this value it can be concluded that the substance can be classified as Aquatic chronic category 2 as per the criteria of CLP regulation.

Toxicity to microorganisms:

In microorganism study conducted according to ISO 8192 "Test for inhibition of oxygen consumption by activated sludge" method for 3 hours duration on activated sludge microorganisms was found to be EC50 at dose concentration > 10000 mg/l.
