Short term toxicity to fish:

Data available for the structurally and functionally similar read across chemicals has been reviewed to determine the short term toxicity of fish of the test chemical Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10-triphenodioxazinediyl) bis[imino-2,1-ethanediylimino [6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2- diy l]]]bis[3-carboxy -, dihydroxide, bis(inner salt) (79771 -28 -1).The studies are as mentioned below:

1)The efficiency of test material was evaluated for its short term toxicity effect on Salmo gairdnerii fish . The toxicity was observed at 24 and 48 h.The effect concentration (LC50) was observed to be 2000 mg/l at 24 h and at 96 h the effect concentration was noted to be 750 mg/l.

2)Test material was subjected to evaluate its acute toxicity effect on Fathead Minnow (Pimephales promelas) 10 Fathead Minnow (Pimephales promelas) were acclimatized in 5-gallon wide mouth glass jar for 10 days before the test in the test temperature 17°C. The test organisms were pre-treated with tetracycline . Test material was subjected in the test vessel upto the concentration of 180 mg/l . No toxic effect were observed after 96 hrs. Hence the effect concentration was considered to be >180g/l .By considering LC50 value it is concluded that test chemical is not toxic tofish and can be considered as “Not classified” according to CLP classification criteria.

Thus, based on the above summarised studies, Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10-triphenodioxazinediyl) bis[imino-2,1-ethanediylimino[6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2-diy l]]]bis[3-carboxy -, dihydroxide, bis(inner salt), hexasodium salt and it’s structurally and functionally similar read across substance, it can be concluded that effect concetration value is in the range 180- 2000 mg/

L. Thus, comparing this value with the criteria of CLP regulation, Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10-triphenodioxazinediyl) bis[imino-2,1-ethanediylimino[6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2-diy l]]]bis[3-carboxy -, dihydroxide, bis(inner salt), hexasodium salt cannot be classified for short term toxicity for fish.

Short term toxicity to aquatic invertebrates:

Data available for the structurally similar read across chemicals has been reviewed to determine the toxicity of aquatic invertebrate of the test chemical Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10 -triphenodioxazinediyl)bis[imino-2,1-ethanediylimino [6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2-diyl]]] bis[3-carboxy-, dihydroxide, bis(inner salt), hexasodium salt (79771 -28 -1).The studies are as mentioned below:

1.Short term toxicity of test material on the growth of daphnia magna. Study was performed for the 24hrs in the static system.Stock solution (100 mg/l and 1 g/l) was stirred during 40 hours before testing. The nominal test concentration were 1, 10, 50 and 100 mg/l, 300, 500, 900 and 1000 mg/l. For the concentration 1, 10, 50 and 100 mg/l 10 daphnids per dose were tested (2 replicates) , 300, 500, 900 and 1000 mg/l with 5 daphnids per dose (4 replicates).

No immobilization were observed . The effect concentration for the immobilization of daphnia magna due to the exposure of test material, was  EC0 was >1000 mg/l.

2.Determination of short term toxicity of test material on the growth of daphnia magna. Test was performed in static system under the proper temperature and pH conditions as per the guidelines.

 No immobilization and abnormal behaviour of the Daphnia was observed in the blank control. At 100 mg/l, after 24 h one Daphnia was immobilized in one jar (5 % immobilisation). After 48 h one Daphnia was immobilized in each jar (10 % immobilisation). Therefore, the EC0 value after 24 and 48 h was below 100 mg/l. The EC50 value after 24 and 48 h could not be determined, but was clearly higher than 100 mg/l.

 

Based on the immobilisation of daphnia magna due to the exposure of test chemical for 48hrs, the EC0 was <100 mg/l and the EC50 was > 100mg/l.

By considering EC50 value it is concluded that test chemical is not toxic to aquatic invertebrate and can be considered as “Not classified” according to CLP classification criteria.

Toxicity to aquatic algae and cyanobacteria:

Data available for the structurally similar read across chemicals has been reviewed to determine the toxicity of aquatic algae and cyanobacteria of the test chemical Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10 -triphenodioxazinediyl)bis[imino-2,1-ethanediylimino [6-[(2,5-disulfophenyl) amino] -1,3,5-triazine-4,2-diyl]]] bis[3-carboxy-, dihydroxide, bis(inner salt), hexasodium salt (79771 -28 -1).The studies are as mentioned below:

1.Evaluations of the effect of test chemical on the growth of Scenedesmus subspicatus. Test was performed according to the standard OECD guidelines in the static system for total 72hrs.

  Based on the growth rate inhibition of Scenedesmus subspicatus by the chemical exposure for 96hrs, the EC50 was > 1000 mg/l, NOEC was 500mg/l and the LOEC was 1000 mg/l. Thus the chemical was consider to nontoxic and cannot be consider to be classified as toxic as per the CLP classification criteria.

2.Evaluations of the effect of test chemical on the growth of Pseudokirchnerella subcapitata. Test was performed according to the standard OECD guidelines in the static system for total 72hrs.

 After the 72hrs of exposure at the test concentration of 100 mg/l a slight inhibition of algae growth (18.4 % for biomass, 5.3 % for growth rate) was observed. Therefore, the NOEC and EC0 was determined to be below 100 mg/l, the EC50 was > 100 mg/l.

 Based on the inhibition of growth rate and the biomass of green algae Pseudokirchnerella subcapitata by the contact with test chemical for 72hrs, the EC50 was > 100 mg/l . Thus the chemical was consider to nontoxic and cannot be consider to be classified as toxic as per the CLP classification criteria.

Toxicity to microorganism:

Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10 -triphenodioxazinediyl)bis[imino-2,1-ethanediylimino [6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2-diyl]]] bis[3-carboxy-, dihydroxide, bis(inner salt), hexasodium salt (79771 -28 -1).The studies are as mentioned below:

1.Activated Sludge, Respiration Inhibition Test was performed for test material according to OECD guideline 209. The effect concentration of test material on activated sludge was observed to be > 1000 mg/l when evaluated after 3 hrs.

2.Test to evaluate toxicity to microorganism was performed for the test chemical . The test chemical was subjected to Pseudomonas fluorescens culture no toxic effects were observed . No inhibition of respiration at a test substance concentration of 1000 mg/l.

No toxic effects of Pyridinium, 1,1'-[(6,13-dichloro-4,11-disulfo-3,10 -triphenodioxazinediyl)bis[imino-2,1-ethanediylimino [6-[(2,5-disulfophenyl)amino] -1,3,5-triazine-4,2-diyl]]] bis[3-carboxy-, dihydroxide, bis(inner salt), hexasodium salt
