- Daphnia magna were cultured in the laboratory. Cultures were maintained in a synthetic medium with the same chemical composition and similar pH dissolved oxygen concentrations as that used to prepare the test solutions.
The cultures are fed daily with cells of the green algae Chlorella vulgaris. Cultures are reset using 10 neonates, when they become gravid their numbers are reduced from 10 to 5 and the production of neoantes results in their daily removal.

- At 48 hours Daphnia exposed to 1.8 mg/L of the test substance and above were not swimming freely. Movement was spasmodic and only observed at the bottom of the test vessels. At a concentration of 1.0 mg/L and below, this abnormal behaviour was not observed.
The % immobile at 48 hours is reported as 0, 10, 0, 0, 0, 60 and 100% at respectively nominal concentrations of 0, 0.56, 1.0, 1.8, 3.2, 5.6 and 10 mg/L.

- The EC50 (48 hours) has been determined to be 5.2 mg/L with 95% confidence limits of 3.2 - 10 mg/L and the NOEC (48 hours) NOEC (48 hours) was determined to be 1.0 mg/L in this test conditions.
- Executive summary:
The acute toxicity of Anther to the freshwater cladoceran Daphnia Magna was determined according to the OECD Guideline 202. The test was conducted under static renewal conditions with test media renewal at 24 hours. Daphnia were exposed to a range of concentrations 0.56 - 10.0 mg/L for a total period of 48 hours. There was no analysis of the test substance during the test period but from its chemical structure the substance is expected to be hydrolytically stable. The EC50s based on nominal concentrations and calculated by the Binomial method are : EC50 (24 hours) > 10mg/L; EC50 (48 hours) = 5.2 mg/L with 95% confidence limits of 3.2 - 10 mg/L. The corresponding NOECs are 5.6 and 1.0mg/L respectively.
At 48 hours, Daphnia in the higher concentrations were classed as mobile but it was considered that these were affected by the test substance since they were observed moving through the lower levels of the medium only in a spasmodic fashion. At a concentration of 1.0 mg/L and below, this abnormal behaviour was not observed.
