- Samples of 10 ml test solution were taken from each prepared solution at the start of test and each test vessel after 48 hours. The samples were analyzed with HPLC. When analysis of test solution was conducted, measurement using standard solution was done for each test sample analysis and the concentration was determined with peak height of the spectrum.

- 1000 mg of sodium p-toluenesulfonate was added to 1L measuring flask and then diluted to 1L with diluting water. Thus obtained solution was used for the test.

These animals originated from National Institute of Environment Study were cultivated in this Laboratory.
Breeding method to prepare young daphnia for the test.
Visually healthy and enough size adult females, which are brood progeny were selected from stock cultivated animals and transfer them to another vessels. In the next day, new born animals were moved to other vessels. These young animals have been bred for 2 or 3 weeks under following conditions to use as parental animals for the test animal.
Young animals were removed at least twice a week from females which were matured and began to deliver offspring. After from 2 to 4 weeks, just before date of start of exposure, brood females in the brood chamber were selected and next day (within 24 hours), delivered young animals were used for the test. However, daphnia from batches, where many dead animals, resting-eggs or male offspring were observed were not used.

Diluting water
Dechlorinated water [Industrial water (drinking water grade treated with chlorine) in the Nishiki plant, Kureha Corporation, is treated with activated carbon to remove residual chlorine and then aeration with air] was used for the test. Major water qualities of diluting water were that hardness was 26mg/L (CaC03 converted) and pH was 7.8.

Determination of test concentration
Based on preliminary test results, 1000 mg/L and control were set.
Test procedure
On the previous day of start of exposure, all young daphnids continuous cultivated for 2 to 4 weeks were withdrawn from vessels.
On the start day of exposure young animals aged less than 25 hours were moved to other vessels and used for the test.
After measured pH, DOC and water temperature of each test solution, 5 of water fleas were added into the test vessel.

Description of key information
The immobilisation of Daphnia magna was determined acording to the OECD 202 test guideline in accordance with GLP, with exposure to Daphnia magana under static conditions at a test concentration of 1000 mg/l. As the substance is highly water soluble and hydrolytically stable no analytical monitoring was conducted. Following a 48 hour exposure, no immobilisation occured. The EC50 is >1000 mg/l.
