According to the results of a pre-experiment, DA 8506 is not soluble at a concentration of 100 mg/L in the test water. Therefore, three WAFs (water accommodated fractions) with loading rates of the test item of 1.0, 10 and 100 mg/L were prepared. Two individual dispersions of the test item with loading rates of 10 and 100 mg/L were prepared by weighing adequate amounts of the test item by means of an analytical balance:
Loading rate 10 mg/L: 50.1 mg suspended in 5000 ml of test water
Loading rate 100 mg/L: 301.8 mg suspended in 3000 ml of test water

The test item was dispersed in the test media using ultrasonic treatment for 15 minutes. Then, the dispersions were stirred by magnetic stirrers at room temperature in the dark over 96 hours to dissolve a maximum amount of the test item in the test media. No auxiliary solvent or emulsifier was used. After the stirring period of 96 hours, each of the dispersions was filtered through a membrane filter (Schleicher & Schuell, Type NC45, pore size 0.45 μm). The filtrates were tested on the fish as WAFs.

The WAF of the lowest loading rate of 1.0 mg/L was prepared by dilution (1: 10) of the WAF with the loading rate of 10 mg/L with test water, due to technical reasons.

The finished test solutions were clear with no visible test material.

In each treatment and the control, 3 fish were tested in about 3 liters of test medium in a glass-aquarium. The mean body length of the test fish at the start of the test was 2.8 ± 0.22 cm (mean ± standard deviation). The mean body wet weight was 0.17 ± 0.03 g. The fish were acclimated to the conditions of the test according to the test guidelines.

Conclusions:
Based on this study, chlorinated methyl esters are not acutely toxic to fish at loading level up to 100 mg/L. This concentration is well above the expected water solubility limit for this substance.
Executive summary:

A 96 hour study of chlorinated methyl esters in Zebra fish was conducted using water accommodated fractions (stirred for 96 hours) at nominal concentrations of 1, 10 and 100 mg/L. There were no effects on mobility of mortality at at concentration. The LC50 is >100 mg/L. The EC0 for this study was 100 mg/L.
