Deviations from the Guideline
The following deviations were observed:
• The stock solutions to be autoclaved were autoclaved for 20 minutes instead of the required 15 minutes. This can be considered uncritical, as the goal of sterility is achieved.
• The pre-culture was prepared 5 days before the start of experiment 1. As all validity criteria were met and exponential growth was observed in the blank control this was stated as uncritical.
• The pre-culture was prepared 5 days before the start of experiment 2. As all validity criteria were met and exponential growth was observed in the blank control this was stated as uncritical.
• The temperature in the experiment 1 was in the range 22.8 – 24.7 °C, and therefore not in the desired range (21.0 - 24.0 °C). As all validity criteria were met and expo-nential growth was observed in the blank control this was stated as uncritical.
• The temperature in the experiment 2 was in the range 21.6 – 25.6 °C, and therefore not in the desired range (21.0 - 24.0 °C). As all validity criteria were met and expo-nential growth was observed in the blank control this was stated as uncritical.
• The temperature during pre-culture incubation for experiment 1 was in the range 23.0 – 27.0 °C, and therefore not in the desired range (21.0 - 24.0 °C). As all validity criteria were met and exponential growth was observed in the blank control this was stated as uncritical.

TREATMENT OF TEST MATERIAL PRIOR TO TESTING
- Treatment of test material prior to testing:
Because of its hygroscopic properties, the test item was dried over night at 140 – 150 °C.
A stock solution containing 101.6 mg/L (first experiment) resp. 100.0 mg/L (second exper-iment) test item in algal medium (demineralised water enriched with minerals but without algae) was prepared. Lower test concentrations were prepared by dilution of the stock solution in algal medium

Sample Preparation
Before measurement the taken samples were diluted, if necessary, to stay in the calibrated range.
Samples with nominal concentration 100 mg/L were diluted tenfold, samples with nominal concentration 32 mg/L were diluted twofold with test medium.

Miscellaneous
On each sampling day, validity of calibration was controlled by measuring QC samples (1 and 10 mg/L). If the deviation was higher than ± 3 % and lower than or equal ± 15 %, the sensitivity correction factor from the QC samples was taken into account
First experiment:
At the beginning of the study a new calibration was performed and the measured QC samples were not taken into account. The mean recovery of the QC samples at the end of the experiment (109.2 % and 105.5 %) were higher than ± 3 % and therefore, taken into account.
Second experiment:
In the second experiment the mean recovery of the QC sample was taken into account at the beginning (92.8 % and 95.5 %), after 24 hours (93.2 %) and at the end of the test (96.5 %).

Details on results:
Significant inhibition of algal growth was observed at all concentrations. However, a clear dose-response relationship was observed.
The content of the test item in the test solutions was determined using HPLC. In the first experiment the content of the test item was determined at the beginning and at the end of the test. In the second experiment the content of the test item was measured daily.
At the lowest concentration 0.32 mg/L no test item was detectable. In the 5 higher concen-trations, at the beginning of the test, the correlation between measured concentration and nominal concentration was very good. At the following measurements a decrease of the measured concentrations could be observed, especially in the lower concentrations. At the end of the test, test item could only be detected in the 2 highest concentrations 32 mg/L and 100 mg/L.
Results with reference substance (positive control):
The 72h-EC50 values of potassium dichromate (K2Cr2O7, CAS No. 7778-50-9) were de-termined in a separate reference test. The values lay within the range of the laboratory (growth rate 0.65 - 1.10 mg/L, yield 0.21 – 0.66 mg/L).

Executive summary:

Findings and Results:

 

Two valid experiments were performed.

In the first experiment the measured values at the end of the test are not usable, because of an error during the analytical measurement or sample preparation. Therefore, the experiment was repeated. Because the test item is not stable at low concentrations, in the second experiment analytical measurements were performed every 24 hours. The biological results were based on the result of the second experiment only.

 

The study was performed using 6 concentrations ranging from 0.32 to 100 mg/L (nominal). Incubation time (test system Desmodesmus subspicatus)was 72 hours. The cell concentration of each replicate was determined by measuring the cell numbers every 24 hours with an electronic particle counter. Growth rate µ and the yield[1]were determined from the cell number at the respective observation times.

Significant inhibition of algal growth was observed at all concentrations. However, a clear dose-response relationship was observed.

 

The content of the test item in the test solutions was determined using HPLC. In the first experiment the content of the test item was determined at the beginning and at the end of the test. In the second experiment the content of the test item was measured daily.

At the lowest concentration 0.32 mg/L no test item was detectable. In the 5 higher concentrations, at the beginning of the test, the correlation between measured concentration and nominal concentration was very good. At the following measurements a decrease of the measured concentrations could be observed, especially in the lower concentrations. At the end of the test, test item could only be detected in the 2 highest concentrations 32 mg/L and 100 mg/L.

The measured concentrations within the exposure period were not within the range of ± 20 % of the nominal values. Therefore, the determination of the results was based on the geometric mean of the measured concentrations (see § 39 OECD 201). The lowest concentration 0.32 mg/L was excluded from evaluation because no test item was detectable at any measurement.

For the treatments 1 mg/L, 3.2 mg/L and 10 mg/L the half of the limit of quantification was used for calculation of the geometric mean because no test item was detectable at the end of the test.


[1]Yield (according to OECD Guideline 201) is defined as the biomass at the end of the exposure period minus the biomass at the start of the exposure period. Calculation see under 8.2 Growth and growth inhibition are quantified from measurements of the algal biomass as a function of time. Algal biomass is defined as the dry weight per volume, e.g. mg algae/L test solution. However, dry weight is difficult to measure and therefore surrogate parameters are used. Of these surrogates, cell counts are most often used.
