Executive summary:

Short term toxicity to Pseudokirchneriella subcapitata (algae) study was carried out for 72 hrs. The study was performed according to OECD Guideline 201 (Alga, Growth Inhibition Test). The study was based on the effects of the test compound on Pseudokirchneriella subcapitatain a static fresh water system. Pseudokirchneriella subcapitata was used as test organism. Analytical monitoring was done in which the NPOC (non-purgeable organic carbon) analysis was performed. Cultured algae were added to the WAF (Water – Accommodated Fraction) and Potassium dichromate was used as positive control in the study. Based on the effect on growth rate and biomass of the test organism Pseudokirchneriella subcapitata, the72hr-ErC50and EbC50 values of test chemical was determined as 0.83 and 0.44mg/L, respectively.Thus, based on EC50 value, it can be concluded that the substance can be considered as toxic to aquatic organisms and thus can be classifiedas aquatic acute category 1 / chronic 1 as per the CLP classification criteria.

Description of key information

1. Based on effect on growth rate of the test organism green algae, the 72 hr EC50 and NOEC value was determined to be 0.34 and 0.026 mg/l, respectively and on the basis of AUG, the 72 hr EC50 and NOEC value was determined to be 0.093 and 0.026 mg/l, respectively.

2. Based on the effect on growth rate and biomass of the test organism Pseudokirchneriella subcapitata, the 72hr ErC50and EbC50 values of test chemical was determined as 0.83 and 0.44 mg/L, respectively.

Based on both studies it was concluded that the Reaction mass of 3H-​Indolium, 2-​[2-​[4-​(dimethylamino)​phenyl]​ethenyl]​-​1,​3,​3-​trimethyl-​ & acetate was toxic and classified as aquatic acute 1 and chronic 1 as per the CLP criteria.

Data available for the structurally similar read across chemicals has been reviewed to determine the toxicity of aquatic algae and cyanobacteria of the test Reaction mass of 3H-​Indolium, 2-​[2-​[4-​(dimethylamino)​phenyl]​ethenyl]​-​1,​3,​3-​trimethyl-​ & acetate. The studies are as mentioned below:

 

In the first study from authoritative database short term toxicity to green algae study was carried out for 72 hrs. The study was based on the effects of the test compound on green algae in a static fresh water system. Based on effect on growth rate of the test organism green algae, the 72 hr EC50 and NOEC value was determined to be 0.34 and 0.026 mg/l, respectively and on the basis of AUG, the 72 hr EC50 and NOEC value was determined to be 0.093 and 0.026 mg/l, respectively.
