In Article 13 of Regulation (EC) No 1907/2006, it is laid down that information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI (of the same Regulation) are met.

 

According to Annex XI of Regulation (EC) No 1907/2006 (Q)SAR results can be used if (1) the scientific validity of the (Q)SAR model has been established, (2) the substance falls within the applicability domain of the (Q)SAR model, (3) the results are adequate for the purpose of classification and labeling and/or risk assessment and (4) adequate and reliable documentation of the applied method is provided.

 

For the assessment of 3,4 -dimethoxyphenethylamine (CAS 120 -20 -7) (Q)SAR results were used for toxicity to aquatic organisms.The criteria listed in Annex XI of Regulation (EC) No 1907/2006 are considered to be adequately fulfilled and therefore the endpoint(s) sufficiently covered and suitable for risk assessment.

 

Therefore, experimental studies on aquatic toxicity are not provided.

Assessment

No experimental data are available on the toxicity to aquatic algae for 3,4 -dimethoxyphenethylamine (CAS 120 -20 -7). Therefore, data have been estimated using ECOSAR v1.11 (US EPA, 2012). In addition, a read-across approach to the structurally analogous substances 3,4 -dimethoxyphenylacetonitrile (CAS 93-17-4), 4 -allylveratrole (CAS 93-15-2) was performed.

ECOSAR calculated the algal toxicity based on user-entered values for log Kow, water solubility and melting point. ECOSAR assigned the substance to the chemical class of aliphatic amines. The 96-h EC50 was estimated to be 575 mg/L, the chronic value was 147 mg/L. The substance was within the applicability domain of the model.

 A study on growth inhibition of the freshwater algae Rhaphidocelis subcapitata, exposed to the analogue 4 -allylveratrole (CAS 93-15-2) for 72h, was performed according to OECD TG 201. Based on measured test concentrations, the 72-h ErC50 was determined to be 22 mg/L (72-h NOEC = 4.6 mg/L). The data are from a reliable source. Though no test report is available at present the data from this source are known to be well documented and tests are performed according to GLP and OECD guidelines (NITE, J-CHECK database, Japan).

The other source chemical 3,4 -dimethoxyphenylacetonitrile (CAS 93-17-4) was tested according to OECD TG 201 using Desmodesmus subspicatus as test species. Test concentration were analytically verified. Based on nominal test concentrations, the 72-h ErC50 was determined to be greater than 100 mg/L. The 72-h ErC10 was 41.8 mg/L (72-h NOEC = 25 mg/L; BASF AG, 2003; report no. 02/0651/60/1).

Based on the available QSAR data the target substance is with high probability acutely not harmful to aquatic algae; however, based on a weight-of-evidence approach and a worst-case assessment, 3,4 -dimethoxyphenethylamine (CAS 120-20-7) is assessed to be acutely harmful to aquatic algae and cyanobacteria. Chronic effects are not expected (NOEC/EC10 > 1 mg/L).
