Since no studies investigating the toxicity to aquatic algae of fatty acids, coco, di-and triesters with trimethylolpropane (CAS 85566-29-6) are available for this endpoint, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 a read across to the structurally related category members Fatty acids, 8-10 (even numbered), di- and triesters with propylidynetrimethanol (CAS 11138-60-6), Fatty acids, C8-10(even), C14-18(even) and C16-18(even)-unsatd., triesters with trimethylolpropane (former CAS 85186-89-6), Fatty acids, C6-18, triesters with trimethylolpropane (CAS 91050-88-3) and fatty acids, C16-18 (even numbered) and C16-18-unsatd. (even numbered), triesters with trimethylolpropane (CAS 68002-79-9) was conducted. The read-across substances cover the lower- (CAS 11138-60-6) and the upper-end (CAS 68002-79-9 and former CAS 85186-89-6) of the substance and thus allow for interpolation to Fatty acids, coco, triester with trimethylolpropane (CAS 85566-29-6). Moreover, Fatty acids, C6-18, triesters with trimethylolpropane (CAS 91050-88-3) covers the whole range from the lower to the upper end of C-chain distribution of the substance. This read-across is justified in detail in the overall summary (IUCLID Section 6.1) and within the category justification in IUCLID Section 13. In this case of read-across, the best suited (highest degree of structural similarity, nearest physico-chemical properties) read-across substance was used for the assessment. Nevertheless, as it can be seen in the data matrix of the category justification in section 13 and the overall summary (IUCLID section 6.1), all reliable data in the category support the hazard assessment ofeach category member by showing a consistent pattern of results. The first study with the read-across substance fatty acids, C16-18 (even numbered) and C16-18-unsatd. (even numbered), triesters with trimethylolpropane (CAS 68002-79-9), was performed according to OECD 201 (GLP) with the green algae Desmodesmus subspicatus. An inhibition of growth rate of 24% after 72 h exposure was observed. Therefore, the ErL50 can be derived as > 100 mg/L (WAF loading rate). The test result shows low toxicities of the test substance far in excess of natural occurring concentrations (water solubility < 0.92 mg/L). Therefore, the test substance is not harmful to algae. The second study (Häner, 2007) with the read-across substance the read-across substance fatty acids, C8-10 (even numbered), di-and triesters with propylidynetrimethanol (CAS 11138-60-6) was performed according to OECD 201 with Desmodesmus subspicatus and determined an ErL0 (72 h) for growth rate of ≥ 100 mg/L and an ErL50 > 100 mg/L (WAF loading rate). The results show no toxicities of the read-across substance fatty acids, C8-10 (even numbered), di-and triesters with propylidynetrimethanol (CAS 11138-60-6) in the range of water solubility. The third study with the read-across substance Fatty acids, C8-18 and C18-unsatd., esters with trimethylolpropane (former CAS 85186-89-6) was performed according to EU Method C.3 (Wierich and Özkabakcioglu, 1999). The static GLP-study with Desmodesmus subspicatus shows an EL50 (72h) > 100 mg/L (WAF loading rate) for the growth rate. Therefore, the substance is not harmful to freshwater algae. The fourth study with the read-across substance Fatty acids, C6-18, triesters with trimethylolpropane (CAS 91050-88-3) was performed according to OECD 201 with the freshwater algae Pseudokirchneriella subcapitata (Bouillon, 2011). Algae were exposed to loading rate of 1000 mg/L (Water Accommodated Fraction) for 72 h. The highest observed algae growth inhibition was 15.5%. Overall, the mean algae growth inhibition was 7.9%. The results show that EL50 (72 h) was > 1000 mg/L.
Based on the results from structurally related read-across substances (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which are characterized by a similar ecotoxicological profile and comparable structure, it can be concluded that fatty acids, coco, di-and triesters with trimethylolpropane (CAS 85566-29-6) will not exhibit effects to aquatic algae up to the limit of water solubility.
