The short term toxicity to fish was determined in accordance with OECD Guideline 203 taking into consideration OECD (2010): No.126 Short Guidance On The Threshold Approach for Acute Fish Toxicity, 2010.
The test item was evaluated in a static limit test, 96 h exposure in treated test medium; 7 organisms ( zebrafish; Danio rerio) for the test concentration and the control were used. The test solution (35 mg/L) was prepared by dilution of the test item in test medium and application of a defined volume of the stock solution to the test vessel. Assessments on effects and mortality after 0, 4, 24, 48, 72 and 96 hours were conducted.
The LC50 (96 h) of the test item was determined to be > 35.0 mg/L test item (nominal). The corresponding NOEC (mortality) (96 h) was 35.0 mg/L test item (nominal). No sublethal effects were observed in the control and the test item concentration of 35.0 mg/L after 96 h. The absence of mortality at the threshold concentration of 35.0 mg/L indicates that fish is not the most sensitive group of test organism and that the LC50 is greater than the limit test concentration.
Daphnia
The acute toxicity of the test substance to aquatic invertebrates (Daphnia magna) was performed in accordance with OECD Guideline 202.
A semi-static main test with nominal concentrations of 100, 50.0, 25.0, 12.5 6.25 mg/L and control was performed using twenty organisms per test concentration (4 replicates of 5 animals each). The duration of the test was 48 hours.
Temperature, pH-value and oxygen concentration of the test solutions measured after 0, 24 hours fresh and 48 hours aged are reported. Hardness of the test water was measured on the day of application. Analytical samples taken at 0 hours (initial value) and 24 hours from fresh and 48 hours aged test solutions were analysed from control and 50.0 and 100 mg/L.
According to the results of the test, the EC50 (48 h) for immobilisation was supposed to be > 100 mg/L (nominal). The corresponding NOEC (48 h) was 50.0 mg/L (nominal).
Algae
The study was performed according to OECD test guideline 201 (2011). Alage ( Pseudokirchneriella subcapitata) were exposed for 72 hours to test concentrations of 100, 31.3, 9.77, 3.05, 0.954 mg/L and control under static conditions. Six replicates were employed for the control and three for each test item concentration. Initial target cell densities of 0.5 × 104 cells/mL were employed for the individual replicates. The increase of cell numbers was assessed over a test period of 72 hours. The test was performed in 100 mL Erlenmeyer flasks each containing ~ 50 mL test solution. The pH was recorded at test start and test end. Temperature was measured continuously over the whole test period and recorded daily. Light intensity of the continuous illumination was measured at test start. Analytical samples were taken and analysed from control and all test item concentrations at 0 hours (initial value) from fresh test solutions and after 72 hours from aged solutions.
The validity critera of the test method were met.
Significant inhibitory effects were determined for growth rate at test item concentrations of 9.77 mg/L (nominal) and above. For yield significant inhibitory effects were determined at 3.05 mg/L (nominal) and above. The overall LOEC was therefore determined to be 3.05 mg/L (nominal), the corresponding NOEC was set at 0.954 mg/L (nominal). The EC10-value for growth rate (ErC10) was determined to be 5.54 mg/L (nominal). The EC10-value for yield (EyC10) was 0.964 mg/L (nominal). The EC20-value for growth rate (ErC20) was determined to be 11.2 mg/L (nominal). The EC20-value for yield (EyC20) was 2.35 mg/L (nominal). The EC50-value for growth rate (ErC50) was determined to be 32.2 mg/L (nominal). The EC50-value for yield (EyC50) was 8.99 mg/L (nominal).
Micro-organisms
An OECD 209 Activated sludge inhibition test (2010) was conducted with the test item. This was to assess the inhibition effect of different concentrations of test item (80.0, 160.0, 288.0, 576.0 and 1037.0 mg/L) to micro-organisms of activated sludge by measuring the respiration rate after adding a standard amount of synthetic sewage feed. The test met all guideline validity criteria.
The results for the inhibition of the total respiration of activated sludge were:
EC50 = 145.4 mg/L (95%-CL: 1.175 – 15679.727 mg/L)
EC10 = 0.603 mg/L (95%-CL: 0.011 – 31.807 mg/L)
LOEC <= 80.0 mg/L
NOEC < 80.0 mg/L
