Description of key information
Not harmful/toxic for aquatic life based on the following:
Short-term toxicity to fish: LD50 (96h, Cyprinus carpio) = 180 - 320 mg/l (nominal)
Short-term toxicity to invertebrates: EC50 (48h, Daphnia magna) > 100 mg/l (nominal; solubility limit)
Long-term toxicity to invertebrates: NOEC (21d, Daphnia magna) ≥ 10 mg/l (nominal), based on mortality and reproduction
Toxicity to aquatic algae: ErC50 (72h, Pseudokirchneriella subcapitata) = 93 mg/l (nominal); NOEC (72h) = 10 mg/l (nominal)
Toxicity to microorganisms: EC50 (3h, aerobic wastewater microorganisms) > 1000 mg/l (nominal)
Additional information
Available data on both Direct Blue 267 and the structural analogue, Similar Substance 01, was considered for the aquatic toxicity assessment and classification.
The use of information on the structural analogue can be considered as suitable and appropriate as the minor structural differences are not expected to influence the ecotoxicological properties; details on the approach followed are included in the document attached to IUCLID Section 13.
Direct Blue 267 is not rapidly degradable; however, based on the low logPow, the substance is expected to possess a low potential for bioaccumulation.
Data is available on Direct Blue 267 for short-term toxicity to Daphnia magna; data on short-term toxicity to fish (Cyprinus carpio), short-term and long-term toxicity to Daphnia magna and toxicity to Pseudokirchneriella subcapitata is available for the structural analogue, Similar Susbtance 01.
Similar Substance 01 resulted not harful/toxic to either fish or algae; the same behavoiur is expected for Direct Blue 267.
Regarding short-term toxicity to Daphnia magna, Direct Blue 267 did not show acute toxic effects up to solubility limit in test water (i.e. 2.1 mg/l). Comparable results, in terms of solubility and effects, were obtained in the experiment performed on Similar Substance 01. In addition, a long-term toxicity test is also availavle, in which the No Observed Effect Concentration (21d) was found to be higher than 10 mg/l (nominal), based on mortality and reproduction.
All experiments results have been considered in order to assess the aquatic toxicity potential of the substance.
In all cases, L(E)C50 values recorded resulted to be higher than 1 mg/l, therefore, the substance does not meet the criteria for classification for acute aquatic hazard. Regarding long-term toxicity, adequate chronic toxicity data are available on Daphnia magna, which indicate the NOEC as higher than 1 mg/l, thus the substance does not meet the criteria to be classified for long-term aquatic hazard.
