Description of key information

Hydrolysis of 2-amino-6-chloro-4-nitrophenol was lower than 10 % for all the three pH values (pH 4, 7 and 9) tested after 5 days at 50°C. As hydrolysis lower than 10 % was observed after 5 days during a preliminary test, so the test item should be regarded as hydrolytically stable.

Hydrolysis of 2-amino-6-chloro-4-nitrophenol was determined under GLP and following OECD 111 and EU C.7 method.

A preliminary hydrolysis test was conducted with buffer solutions of pH 4.0, 7.0 and 9.0 at temperature of 50°C. Two sample solutions were prepared for each pH buffer by adding 1.00 mL of a 20 mg/mL test substance solution in methanol (HPLC grade) into a 100 mL buffer solution.

Samples were collected at 0 hour (test start), after 2.4 hours and 5 days (after placing the sample solutions into the water thermostatic bath at 50°C). The active ingredient content of the solutions was analyzed by UV/Visible detector, with internal standard. 

Hydrolysis of lower than 10 % was observed for all the three pH conditions tested after 5 days at 50°C. So the test itel is considered as hydrolytically stable.

As per recommendations of the OJEC C.7 method, if the preliminary test indicates that 50% or more of the test substance has been hydrolyzed in 2.4 hours at 50°C or less than 10% has been hydrolyzed after five days at each of the three pH values (4, 7 and 9), no further testing is necessary. Therefore, no definitive test was performed.

This hydrolysis test is classified as acceptable, and satisfies the guideline requirements of the OECD 111 guideline.
