Test organisms

Test organisms (species):
activated sludge of a predominantly domestic sewage
Details on inoculum:
- Name and location of sewage treatment plant where inoculum was collected: Sewage sludge for a municipal STP
- Preparation of inoculum for exposure: The sewage sludge was treated by centrifugation and washing with drinking water (4x times).
- Initial biomass concentration: 3,5 g dry weight/L (1.4 g dry weight/L per test vessel)

Study design

Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
3 h

Test conditions

Test temperature:
18-22 °C, test start 18-18.5 °C, during O2 measurement 19-20 °C
pH:
8.1
Dissolved oxygen:
24.32-24.68 mg O2/ L*h
Nominal and measured concentrations:
20 mg/L, 41.2 mg/L, 85.8 mg/L, 133 mg/L, 262.2 mg/L
Details on test conditions:
100 mL dest. water was added to the defined amount of test substance. 16 mL synthetic sewage water and 200 mL of the inoculum were added to the test substance and the volume was filled up to 500 mL with drinking water.
After 3 hours, the pH value has been determined and O2 consumption has been measured for 8 min.

Results and discussion

Effect concentrations
Key result
Duration:
3 h
Dose descriptor:
EC50
Effect conc.:
> 260 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
inhibition of total respiration
Remarks on result:
other:
Remarks:
The test substance revealed no inhibition on the respiration of sewage sludge.
Details on results:
In the tested concentration range of 20 to 262.2 mg/L test substance, no inhibition on the respiration of sewage sludge by the test substance has been observed. Hence, a EC10 or an EC50 could not be determined.
Results with reference substance (positive control):
3,5-Dichlorophenol has been tested with a concentration of 5, 7, and 10 mg/L and an EC50 of 6.6 mg/L was measured.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
In the tested concentration range of 20; 41,2; 85,8; 133 and 262,2 mg / L, neither EC10 nor EC50 was achieved. The test substance did not have an inhibitory effect on the respiration of the activated sludge.
