220 mg of NaBH4, in 10 ml 1M NaOH were added drop by drop with stirring to 3 g of 4-isobufylacetophenone dissolved in 10 ml of methanol (17 mmol). Then, the reaction mixture was allowed to stand at room temperature for 48 h. After evaporating the methanol, the mixture was
extracted with ethyl acetate and the extract purified by silica PLC using CHCI, as eluent.
Yield 70%, viscous oil.

Results and discussion
- Details on results:
- Hemolysis
- The test substance produced a massive cell lysis within the first minutes after addition at 2.5 x 10^-3 M to the erythrocytes suspension (15 x 10^6 cell/ml) at 37°C.
- Lysis of cells was clearly evident after 2 min at 2.5 x 10^-3 M and after 20 min at 1x10^-3 M. Lower concentration (5x10^-4) were also toxic, but their effects were observed only after longer exposure times.
- There was no lysis in the controls (DMEM + 1% acetone).
Enzyme leakage of cultured fibroblasts.
The test substance was added to culture plates at different concentrations, and the activity of enzymes LDH and GOT was periodically assayed in the culture supernatants. At the end of the experiment, cell viability was also estimated.
- After a few minutes, cells exposed to 2.5 x 10^-3 M were dead.
- Fibroblasts exposed showed extensive LDH leakage after 4h incubations at 1 and 2.5 x10^-3 M (not observed for 5 x 10^-4 M)
- GOT activity in the supernatant was increased after a 4h test substance treatment at 1 and 2.5 x10^-3 M (not observed for 5 x 10^-4 M).
Overall, cytotoxic effects - assayed using the red-blood cell lysis test and the enzyme leakage (LDH and GOT) from cultured fibroblasts - were observed for the test substance 1-(4-isobutylphenyl)-ethanol in both system at concentrations above 500 µM.
