- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: The test concentration was prepared by diluting an acetonic stock solution. The stock
solution will be prepared by weighing 0.00507 g test substance and diluting it in 100 mL
acetone (SI-, stock solution No 1). A second dilution was done: 1 mL SLI was added to
100 mL acetone (SL-2). The required amount (10 ml) of the acetonic stock solution (SL-2) was
given into 100 mL volumetric flasks. For the dilution steps from SL3 to SL6 each time 25 ml
of the corresponding SL solution was added to 50 ml acetone. After evaporating of the
acetone the volumetric flasks were fiIIed with test medium. The test substance was stirred in
M4 medium for about one day at 20 ± 2 0C. The stock solution appears colourless-clear.
