- SAMPLE PREPARATION
- Range finding test: Because of the poor solubility in water of the test item an extraction has been performed to extract the soluble components of the test item in water. The test sample has been prepared at 0.1 g/L in culture medium. The solution has been put under stirring for 48 hours at 10 rpm at room temperature. After 3 hours of settling down the supernatant has been used for the test.

- MEDIUM PREPARATION
25 mL of solution A, B, C, and D have been taken to 1000 mL volume with deionised water. Solution A: 11.76 g/L CaCl2 2H2O; Solution B: 4.93 g/L MgSO4 7H2O; Solution C: 2.59 g/L NaHCO3; Solution D: 0.23 g/L KCl
TREATMENT
4 replications with tested concentrations have been prepared, afterwards Daphnids have been added to the closed vessels (500 mL glass beakers) of the assay sample (5 animals per beaker). The control group has been treated in the same conditions without adding the test item. Daphnids have been kept without light, aeration and feeding for the whole length of the assay. Animals have been kept under observation for a period of 48 hours. An extra replica of each concentration, without organisms, has been performed to evaluate possible loss by evaporation of the sample during the test.
OBSERVATIONS
After 24 and 48 hours from the beginning of the test, the immobilisation of the Daphnids has been evaluated verifying if the organisms, after light stirring of the vessels, were not capable to move in the following 15 seconds. pH, dissolved oxygen have been measured at the beginning and at the end of the test (after 48 hours) in the highest concentration in the treated group and in the control. Temperature of the assay environment has been measured at the beginning and at the end of the test (after 48 hours).
VALIDITY CRITERIA
The immobilisation of control animals must not be higher than 10% at the end of the test. The concentration of dissolved oxygen in the vessels ought to be higher than 3 mg/L during the assay. However, in any case, the concentration of dissolved oxygen must not be lower than 2 mg/L. pH must not change more than 1.5 units - Key result

In a short-term toxicity test to aquatic invertebrates, performed according to OECD Guideline 202 and following GLP, Daphnia magna were exposed to the test item. During the range finding test, the organisms were exposed to 5 different dilutions (0.1, 1, 10, 100, and 1000 mg/L) of the test item for a total period of 48 hours. After the results of the range finding test, an definitive test has been performed. The Daphnia were exposed to 5 dilutions of the test item for 72 hours. The nominal concentrations were 4.26, 9.39, 20.66, 45.45, and 100 mg/L and the measured concentrations were 1.86, 4.08, 8.83, 19.66, and 42.19 mg/L and 1.88, 4.19, 9.16, 20.07, and 49.39 mg/L at T=0 and T=48 hours, respectively. Daphnids have been kept under observations for a period of 48 hours and the number of immobilised organisms and/or possible abnormal behaviours both in control and in treatment groups vessel have been observed. Tests were performed in closed vessels. Dissolved oxygen was 4.90 - 5.09 and 4.31 - 4.85 at the beginning and at the end of the test period, respectively. The pH was 7.78 - 7.81 and 7.77 - 7.84 at the beginning and at the end of the test period, respectively. The test temperature ranged from 20.1 - 20.3°C. Stability of the test item in the definitive test has been determined using gas chromatography technique equipped with mass detector and the test sample was considered stable because the measured concentration of the test item at the end of the test has been maintained within ± 20 % of the concentration at the beginning of the test. The EC50 was determined to be 3.479 mg/L
