Data investigating the aquatic toxicity ofNeopentyl glycol dicaprate (CAS No. 27841-06-1)are not available. Read-across to the structurally related substances Fatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3) and Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5) was conducted for short-term toxicity to fish and toxicity to aquatic algae. Fatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3) was used as source substance for short-term toxicity to aquatic invertebrates. Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5) was used as source substance for long-term toxicity to aquatic invertebrates and toxicity to microorganisms. Based on the high degree of similarity between the structural and physico-chemical properties of the target and source substances, the source substances are considered suitable representatives for the assessment of the aquatic toxicity of the target substance.This read-across approach was applied in order to fulfil the standard information requirements according to Annex VIII of Regulation (EC) No 1907/2006 and is in compliance with Annex XI, 1.5, of Regulation (EC) No 1907/2006.

A detailed analogue approach justification is provided in the technical dossier (please refer to IUCLID Section 13).

For the endpoint short-term toxicity to fish,read-across to the structurally and chemically closely related source substances Fatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3) and Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5)was conducted. Both studies were performed under GLP and according to OECD Guideline 203. For source substanceHeptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5), the LC50 (96 h) was > 0.086 mg/L (measured, geometric mean;Oncorhynchus mykiss).For source substance Fatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3), theLL50 (96 h) was > 100 mg/L (nominal;Oncorhynchus mykiss).No effects were observed up to the limit of water solubility.

For the endpoint short-term toxicity to aquatic invertebrates, a read-across to the structurally related source substanceFatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3)was conducted. TheEL50 (48 h) was determined as > 100 mg/L (WAF, nominal).The study was performed according to OECD Guideline 202. GLP was not applied. No effects were observed up to the limit of water solubility.

For the endpoint long-term toxicity to aquatic invertebrates, a read-across to the structurally related source substanceHeptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5)was conducted.The NOEC was determined as ≥ 0.0019 mg/L loading rate (measured, TWA).The study was performed under GLP and according to OECD Guideline 211. No effects were observed up to the limit of water solubility.

For the endpoint Toxicity to aquatic algae and cyanobacteria, read-across to the structurally related source substancesFatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3) and Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5)was conducted. Both studies were performed under GLP and according to OECD Guideline 201. For source substanceFatty acids, C8-18 and C18-unsatd., esters with neopentyl glycol (CAS 85186-86-3), the EL50 (72 h) was determined as > 100 mg/L (nominal;Desmodesmus subspicatus). The EL0 (72 h) was100 mg/L (nominal). Forsource substanceHeptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5), the EL50 (72 h) was determined as > 0.0065 mg/L (measured, geometric mean;Pseudokirchneriella subcapitata). The NOELR (72 h) was0.0065 mg/L (measured, geometric mean).No effects were observed up to the limit of water solubility.

For the endpoint Toxicity to microorganisms, a read-across to the structurally related source substanceHeptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5)was conducted.The NOEC (3 h) was determined as1000 mg/L (nominal; total respiration). The study was performed under GLP and according to OECD Guideline 209. No effects were observed up to the limit of water solubility.

Based on the available results from structurally related read-across substances (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5), which is characterized by a comparable structure and chemistry, it can be concluded that the target substance Neopentyl glycol dicaprate (CAS No. 27841-06-1 will exhibit comparable effects on aquatic organisms.
