A set of six reference substances; acetanilide, methyl benzoate, naphthalene, 1,2,3 trichlorobenzene, phenanthrene and DDT, were used to calibrate the chromatography system. Individual standard solutions of each were prepared along with a mixed standard containing all six reference substances and formamide. In addition, a solution of formamide was also prepared, to be used as the void volume marker, in order to determine the system dead time.

Two series of injections were made, each consisting of duplicate injections of the test substance, duplicate injections of the reference substances and triplicate injections of formamide (the void volume marker). Injections were made in the following order:
Formamide, Acetanilide standard, Methyl benzoate standard, Naphthalene standard, Test substance, Mixed standard, Test substance, 1,2,3-Trichlorobenzene standard, Phenanthrene standard, DDT standard and Formamide

The reference substances, formamide, and test substance stock solutions were prepared in methanol and these were then diluted with 55:45 v/v methanol:double-distilled water diluting solvent (methanol, 110 mL mixed with double-distilled water, 90 mL) to produce working solutions.

There was good correlation with all the calibration substances for the Log K vs Log Koc graph, with correlation coefficients of 0.9906 in both tests.
Three peaks were attributable to Adoxal, with adsorprtion coefficients of 3.32, 3.57 and 3.86. From the chromatograms, the latter is the major peak. The smaller peaks may actually be those of the impurities present in Adoxal. The test item had a purity of 90.2%.

Testing was performed at 25°C (column temperature), however the values can be considered to represent the values at an unspecified, probably ambient temperature since no mention is made in the guidelines of the temperature at which the tabulated reference substances log Koc values were determined. Adsoprtion coefficients of the test and reference substances are assumed to vary with temperature in the same way.
