Description of key information
2-Ethylhexanoyl chloride is corrosive to the skin. This implies the possible risk of severe eye damage despite the substance was only mildly irritation to the eyes in animal testing.

In a guideline study (OECD TG 404), undiluted 2-ethylhexanoyl chloride (0.5 mL) was applied to the clipped intact skin of New Zealand White rabbits (3/sex) under semi-occlusive conditions for 4 hours (Stillmeadow, 1986). The animals were scored for erythema and eschar formation, edema formation and any other dermal effects or irritation at 1, 24, 48, and 72 hours and 4, 7, 10, 14, 17 and 21 days after removal of the patch. Erythema and oedema were present at each observation time through Day 21, indicating the substance is corrosive to skin.
Eye
0.1 mL 2-ethylhexanoyl chloride was placed into the conjunctival sac of the right eye of each of 9 rabbits (strain not specified;Stillmeadow 1986). The lids were held together for one second. Three of the nine treated eyes were each washed with water for 1 minute, 30 seconds after treatment. The untreated left eye served as a control. Positive staining (with 0.2% fluorescein sodium ophthalmic solution) occurred in four unwashed eyes, which resolved by 48 hours. Positive staining did not occur in any of the washed eyes. 2-ethylhexanoyl chloride was rated mildly irritating.
Justification for classification or non-classification
According to the results of the skin irritation study 2-ethylhexanoyl chloride is classified as skin corrosive cat. 1B (H314) according to Annex I of Directive 1272/2008, this implies the possible risk of severe eye damage.
Based on the skin corrosion and the inhalation toxicity of the substance the additional labeling with EUH071 (corrosive to the respiratory tract) is recommended.
