Sampling and analysis
- Analytical monitoring:
- yes
- Details on sampling:
- Samples were analyzed with a HPLC-DAD method, implemented under non-GLP and documented finally in the GLP raw data.
All concentration levels and the control were analytically verified via HPLC-DAD at the start (0 hours) and at the end of the exposure (72 hours) with algae.
At the start, samples were taken from one additional replicate of each test item concentration and the control and at the end samples were taken from pooled replicates.
Test solutions
- Vehicle:
- no
- Details on test solutions:
- A stock solution of 80.0 mg test item/L was freshly prepared with dilution water (see Table 2). The test solution was treated with ultrasound for 5 minutes at room temperature. The stock solution was checked via laser beam (Tyndall effect) for undissolved test item. The Tyndall effect was negative.
Six concentration levels were tested in a geometrical series with a dilution factor of 2.5: 0.819 - .05 - 5.12 - 12.8 - 32.0 - 80.0 mg/L, corresponding to the geometric mean measured test item concentrations of 0.394 – 1.06 – 2.71 – 6.98 – 20.2 – 58.9 mg/L. The dilution levels are based on the results of a preliminary range finding test (non-GLP).
