- Details on results:
- 1) Test de toxicité aiguë par voie orale chez le rat.
300 mg/kg < DL50 orale < 2000 mg/kg
Un premier traitement avec la substance pure a été
administré à 3 animaux à la dose de 2000 mg/kg. Après 30
minutes, les 3 animaux ont présentés une accélération de la
respiration et une léthargie, 2 animaux salivaient. Après 4
heures la léthargie a persisté chez 2 animaux qui sont morts
dans les 24 heures. Un deuxième traitement a été appliqué à
3 animaux puis 3 autres pour confirmer (6 animaux au
total), à la dose de 300 mg/kg. Aucune anomalie et aucun
décès n'ont été constatés.
Le poids des animaux testés n'a pas changé de façon
significative pendant l'essai, et les résultats de
l'autopsie n'ont révélés aucune anomalie à la fin de
l'étude.
2) Test d'irritation cutanée chez le Lapin : 3 lapins ont
été rasés 24 heures avant le début de l'étude et ont reçu
sur 6 cm² de peau du flanc droit 0,5 mL de substance
appliquée en pansement semi-occlusif pendant 4h. Les animaux
ont été observés à 24, 48 et 72h après les 4 heures
d'application et les scores des œdèmes et érythèmes cotés.
Le flanc gauche a servi de témoin négatif. Les scores des
érythèmes et œdèmes étaient de 0 à tous les stades
d'observation. La moyenne des scores fut donc de 0 pour les
œdèmes et érythèmes.
La substance a été considérée comme non irritante pour la
peau chez le Lapin.
3) Test d'irritation oculaire chez le Lapin : 3 animaux ont
reçus 0,1 ml de substance dans l'œil droit. L'œil gauche non
traité sert de contrôle.
Après 1, 24, 48, 72 heures, 7, 14 et 21 jours les effets
d'irritation sur la cornée, l'iris et la conjonctive ont été
côtés, pour chaque animal. Les moyennes sur chaque animal
pour chaque effet sur 24 h + 48 h + 72 h ont montrées un
effet légèrement irritant, mais la substance est non classée
selon les critères de la Directive 67/548/CE.
4) Test de sensibilisation maximisation sur cobayes : un
essai préliminaires avec 5 animaux a été réalisés pour
déterminer la concentration minimale irritante et la
concentration maximale non irritante pour les phases
d'induction et de challenge. Dans l'essai principal, 20
cobayes ont été traités et 10 animaux ont servis de témoins
négatifs.
Pour les animaux traités :
- Induction intradermique au jour 1 à la concentration de 6%
dans de l'huile de maïs
- induction topique au jour 7 à 100% en pansement occlusif.
- challenge au jour 21 par application topique de la
substance à 100% en pansement occlusif.
24h et 48h après la phase challenge, aucune réaction cutanée
(œdème, érythème) n'a été observée chez les animaux traités
comme chez les témoins indiquant que la substance n'est pas
sensibilisante.
5) Test d'Ames simplifié : 2 souches de Salmonella
typhimurium TA 98 et TA 100, ont été testées avec et sans
système d'activation métabolique à différentes concentration
de substance dans du DMSO : 5,72 ; 1,86 ; 0,64 ; 0,21 ;
0,07 ; et 0 mg/boîte.
Les concentrations ont été déterminés à l'issu d'un essai
préliminaire sur la souche TA 100.
Des contrôles positifs ont été utilisés pour chaque essai.
Aucune cytotoxicité n'a été observée et aucune augmentation
significative du nombre de révertants à toutes les
concentrations testées avec et sans système d'activation
métabolique.
La substance est donc considérée comme non mutagène sur les
deux souches de Salmonella typhimurium TA 98 et TA 100 selon
le test d'Ames.
