- - Sampling method: At the end of the exposure period, the replicates with algae were pooled at each concentration before sampling, except for the 18 mg/L concentration where all three replicates were sampled individually.
Compliance with the quality criteria regarding maintenance of actual concentrations was checked by running a test vessel at an intermediate item concentration but without algae and samples for analysis were taken at the start and at the end of the test period.
Additionally, reserve samples of 1.4 mL were taken from all test solutions for possible analysis. If not already used, these samples were stored in a freezer for a maximum of three months after delivery of the draft report, pending on the decision of the sponsor for additional analysis.

- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Preparation of test solutions started with the highest test concentration of 100 mg/L. No special treatment other than vigorous shaking was needed to fully dissolve the test item in the test medium. The pH was adjusted with HCl (1 mol/L, Merck, Darmstadt, Germany) from 9.6 to 7.1 and from 9.4 to 7.9 in the combined limit/range-finding and the final test, respectively. Lower test concentrations were prepared by subsequent dilutions of the highest concentration in test medium. All final test solutions were clear and colourless.
After preparation, volumes of 50 mL were added to each replicate of the respective test concentration. Subsequently, 1 mL of an algal suspension was added to each replicate providing a cell density of 10^4 cells/mL.

- A 72-h growth inhibition test with the unicellular green alga Pseudokirchneriella subcapitata was performed wit the test substance T000425 according to the OECD guideline 201 (GLP conditions). The measured concentrations were in agreement with the nominal concentrations throughout the test period (93-104%). Based on these results, effect concentrations were expressed in terms of
analytically confirmed nominal concentrations. The 72-h EC50 based on growth rate was determined to be 68 mg/L, the NOEC based on growth rate was 10 mg/L. The results of the test can be considered reliable without restrictions.
