Endpoint:
long-term toxicity to aquatic invertebrates
Type of information:
experimental study
Adequacy of study:
key study
Reliability:

Rationale for reliability incl. deficiencies:
comparable to guideline study with acceptable restrictions
Qualifier:
according to guideline
Guideline:
other: OECD 202, Part II
Deviations:
no
GLP compliance:
not specified
Specific details on test material used for the study:
MCPP-DMA
Purity: 91.6 %
Analytical monitoring:
not specified
Vehicle:
no
Details on test solutions:
The test solution was prepared by dissolving MCPP-P acid in a dimethylamine (DMA) solution. On the day of the experiment, the MCPP-P DMA salt was diluted into the test medium. MCPP-P DMA was used in the nominal concentrations [calculated as amount of MCPP-P acid]: 2.5, 7.4, 22.2, 66.7 and 200 mg/L.
Test organisms (species):
Daphnia magna
Details on test organisms:
The daphnids were fed with the algae Scenedesmus sp.
Test type:
semi-static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
21 d
Hardness:
no data
Test temperature:
20 °C
pH:

Dissolved oxygen:
no data
Salinity:
NA
Conductivity:
no data
Nominal and measured concentrations:
Nominal concentrations: 2.5, 7.4, 22.2, 66.7 and 200 mg/L
Details on test conditions:
The study was performed in glass beakers each containing 200 mL reconstituted water. Ten daphnia, less than 24 hours old were exposed for 4 days in duplicate. The daphnids were then separated each in 80 mL beakers and exposed for 21 days at 20 °C and 16 hours light/day (500 -2000 Lux). The test medium was renewed 3 times per week.
Reference substance (positive control):
no
Key result
Duration:

Effect conc.:
22.2 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
reproduction

A dose-related increase in mortality of adult daphnids was observed for test concentrations 66.7 and 200 mg/L. After 48 hours, no mortality was observed at any concentration. After 96 hours 25 % were dead at 66.7 mg/L and 25 % at 200 mg/L using 2x10 daphnids. The separation in 10 single survivors into brooding beakers (10x1), revealed 20 % mortality at 66.7 mg/L and 100 % at 200 mg/L after 16 days. The reproduction started after 12 days of exposure.

The study was performed with MCPP-P DMA but the result was caculated as amount of MCPP-P acid.

Validity criteria fulfilled:
yes
Conclusions:
For MCPP-P acid, the hydrolysis product of MCPP-P 2-EHE, a 21d-NOEC of 22.2 mg/L was determined for toxicity to reproduction on Daphnia magna.
Executive summary:

In a semi-static reproduction toxicity test, according to OECD Guideline 202 Part II, for MCPP-P acid a 21d-NOEC of 22.2 mg/L was obseved.

Additional information

The long-term toxicity to aquatic invertebrates was investigated with MCPP-P IOE due to structural similarities (read-across).

In a semi-static reproduction toxicity test, according to OECD Guideline 202 Part II, for MCPP-P acid a 21d-NOEC of 22.2 mg/L was obseved.
