Details on test animals or tissues and environmental conditions:
Body weight: 2.3 to 2.55 kg each, 4-5 months of age.

Test system

Vehicle:
unchanged (no vehicle)
Amount / concentration applied:
0.1 ml or 0.1 g.
Observation period (in vivo):
1, 24, 48 and 72 hours and 21 days
Number of animals or in vitro replicates:

Details on study design:
3-Cyanopyridine was tested for eye irritant properties in rabbits. 0.1 g was introduced into the right conjunctival sac of 3 tested animals. Evaluations were made at 1, 24, 48 and 72 hours after application, according to the method of Draize. Evaluation of score is as follows: 1-10 Not irritating; 11-25 Slightly irritating; 26-56 Moderately irritating; 57-110 Severely irritating.

Any other information on results incl. tables

Cornea: turbidity translucent with easily recognizable on region, district or opalescent opaque cornea, the entire cornea was affected in each area, sprouting of vessels in the cornea

Iris: Short term no reaction to light and moderate hyperaemia surrounding the cornea.

Conjunctiva: Diffuse, flesh-colored redness, all sections bloody sometimes, significant swelling with partial eversion of the eyelid

Other changes in the eye area: Mild serous hypersecretion, loss of hair around the eye

The reaction of the cornea after 21 days was not yet completely subsided.

The calculated irritation index amounts to 65.

Applicant's summary and conclusion

Interpretation of results:
Category 1 (irreversible effects on the eye)
Remarks:
Migrated information Criteria used for interpretation of results: EU
Conclusions:
3-Cyanopyridine was tested for eye irritant properties in rabbits after a single exposure. The results indicate strong irritating properties of the rabbit eye.
