Even though the column temperature was 40 °C, this temperature is without influence on the result of the determination. The calibration was done referring to Koc- values of reference substances determined at 25 °C.

Preparation of the test solutions.
- The dead time marker sodium nitrate was dissolved in acetonitrile / demineralized water 9:1(v/v) and diluted with acetonitrile.
- The calibration substances were dissolved and diluted with acetonitrile.
- The test item was dissolved in acetonitrile / demineralized water 1:1(v/v) and diluted with acetonitrile.
- The concentration for the calibration substances was approximately 0.05 to 0.3 mg/mL
- The concentration for the test item was approximately 0.16 mg/mL

Executive summary:

The adsorption coefficient (Koc) of Bayscript gelb GGN was determined according to the HPLC method at 40 °C. In order to comply with the guideline OECD 121 and considering environmental relevance, pH 6 was chosen as pH value for the buffer.

The reference Koc-values in the OECD-guideline presumably refer to 25 °C. Even though the column temperature was 40 °C, this temperature is without influence on the result of the determination. The calibration was done referring to Koc- values of reference substances determined at 25 °C. For the determination of the adsorption coefficient (Koc) six calibration standards with log Koc from 1.45 – 2.75 are established in the EG-test method.

The test item Bayscript gelb GGN consists of two organic parts as an ion pair compound. The compound Benzenesulfonic acid, 3,3’-(carbonylbis(imino-(3-methoxy-4,1-phenylene)-2,1-diazenediyl)) (CAS 25712-08-7, EC 247-203-2) could be determined by HPLC.

The compound 2,2’-Iminobis(ethanol) (CAS 111-42-2, EC 203-868-0) was eluated by HPLC within the dead time (proofed by HPLC and reported in GLP Study “ Partition Coefficient “ number 2016/0068/07).
