Description of key information
The acute toxicity of the read across substance Reaction products of γ-butyrolactone, naphthalene-1,6-diol and phthalic anhydride (EC 946-798-2) to aquatic invertebrates was determined according OECD 202 under GLP conditions. The 48-h EL 50 was determined to be > 100.0 mg/L.
Key value for chemical safety assessment
Additional information
No data on toxicity to aquatic invertebrates were available for the substance itself. Therefore, a read across to the close structural analogue Reaction products ofγ-butyrolactone, naphthalene-1,6-diol and phthalic anhydride (EC 946-798-2) was conducted.
Key study
To assess the toxicity of the close structural analogue (EC 946-798-2) to Daphnia magna (STRAUS), an Acute Toxicity Test according to OECD Guideline 202 was carried out in compliance with GLP principles (BASF SE, 2017). The test was conducted under semi-static conditions over a period of 48 hours with the only (limit) concentration of 100 mg/L. In addition, a negative control (without the test item) and a positive control with the reference substance potassium dichromate was performed. Since the test item is a mixture which has a low water solubility, a Water Soluble Fraction (WSF) was prepared. The WSF and the negative control were analytically verified via UPLC-MS/MS in fresh media at the start of the exposure and at renewal (0 and 24 hours) as well as in 24-hours old media at renewal and at the end of the test (24 and 48 hours). The measured concentrations of the test item were 0.0356 and 0.0421 mg test item/L in fresh media (0 and 24 hours) and 0.0187 and 0.0335 mg test item/L in 24-hour old media (24 and 48 hours). Twenty daphnids (divided into 4 replicates with 5 daphnids each) were exposed to the WSF of the test item and the control. Since the close structural analogue contains a large proportion of unidentified by-components, it was treated as a mixture and the results are based on loadings rather than on mean measured concentrations. All effect values are given based on the nominal loading of the test item of 100 mg/L. In result, at the WSF with a nominal loading of 100 mg/L of the test item, no effects on Daphnia magna were observed. Therefore, the 48-h EL50 of the close structural analogue is considered to be > 100 mg/L. The validity criteria of the test guideline were fulfilled.
