1. HYPOTHESIS FOR THE ANALOGUE APPROACH
This read-across is based on the hypothesis that the source and the target substance have very similar physicochemical and (eco)toxicological properties because their chemical structures are nearly identical. An analogue approach has thus been employed. The target substance is Reactive Red 24 carrying a methyl group, while the source chemical is Reactive Red 24:1 carrying an ethyl group.
The presence of sulphonate groups make both dyes highly water soluble and therefore less critical for human health and environmental issues. Based on their chemical similarity, similar properties are expected in both humans and the environment.

The source substance was tested in a concentration of 30 mg/l. The reference substance (Sodium benzoate) was tested in a concentration of 100 mg/l. Based on the results of the study, biodegradation of the test substance after 28 days was 12 %. The structurally related target substance will show similar behaviour.

Stock solution A:
8,50 g of potassium acid phosphate, 21,75 g of di-potassium hydrogen phosphate, 22,18 g of di-sodium hydrogenphosphate dihydrate and 1,70 g of ammonium chloride were dissolved in distilled water and adjusted to 1 litre. The pH value of this solution was 7,2.

Stock solution B: 22,50 g of magnesium sulfate heptahydrate were dissolved in distilled water and adjusted to 1 litre.
Stock solution C: 36,40 g of calcium chloride dihydrate were dissolved in distilled water and adjusted to 1 litre.

This test method was based on the following conditions:
- test chemicals as sole organic carbon sources
- no adaptation of micro-organisms to the test chemicals
Tested chemicals were inoculated in the testing vessels with micro-organisms. During the test period, the Biochemical Oxygen Demand was measured continuously by means of a BOD-meter. Biodegradability was calculated on the basis of the theoretical oxygen demand (ThOD) or less satisfactorily COD, if the ThOD could not be calculated. Optionally, primary biodegradation could be determined by analysis of dissolved organic carbon (DOC) at the beginning and the end of the test.

Additional information

The target substance is a reactive dye and as such the substance should not be considered to be biodegradable at all. The key study was performed according to OECD TG 302B, inherent biodegradation of the source substance after 28 d was 12%, confirming that the substance should not be considered as inherently or readily biodegradable under the conditions of this test. In addition determination of BOD and COD values and calculation of the BOD5/COD quotient resulted in a value of zero. Based on this the read-across source substance is regarded to be not biodegradable at all. Therefore, the target substance is also considered to be not biodegradable what is expected for a reactive dye.
