In Article 13 of Regulation (EC) No 1907/2006, it is laid down that information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI (of the same Regulation) are met.

According to Annex XI of Regulation (EC) No 1907/2006 (Q)SAR results can be used if (1) the scientific validity of the (Q)SAR model has been established, (2) the substance falls within the applicability domain of the (Q)SAR model, (3) the results are adequate for the purpose of classification and labeling and/or risk assessment and (4) adequate and reliable documentation of the applied method is provided.

The criteria listed in Annex XI of Regulation (EC) No 1907/2006 are considered to be adequately fulfilled and, therefore, the endpoint(s) sufficiently covered and suitable for risk assessment.

 

Assessment

The biodegradability of N-(2-aminoethyl)-1,3-propanediamine was assessed in a GLP guideline study according to OECD 301B (BASF SE, 2014, Rep. no. 22G0210/14G041) with activated non-adapted sludge from domestic sources. Results revealed a degradation of the test substance of 15% (CO2 evolution) after 28 d. In a supporting study conducted with both non-adapted and adapted activated sludge according to OECD 301 F (BASF AG, 1991, Rep. no. 1/91/0407/26/1 (non-adapted) and Rep. no. 1/91/0407/26/2 (adapted)), degradation of the test substance was found to be up to 60% (CO2 degradation) after 35 days (non-adapted). However, it should be noted that sufficient degradation was only observed in one of seven replicates.

The biodegradation of 1,4-Diazabicyclo[2.2.2]octane in water was also estimated using the QSAR method of CATALOGIC 301C v12.17 (OASIS Catalogic v5.15.2), which predicted an O2-consumption (BOD) of 10% after 28d (BASF, 2022).

Based on the available information on the test item N-(2-aminoethyl)-1,3-propanediamine, it can be concluded that the substance is not readily, but moderately/partly biodegradable.
