Preliminary visual estimation of the n-octanol solubility

Appearance of the test mixtures in the preliminary test

Amount of test item in
mg
Total volume in
ml
Appearance of mixture
100 1 dissolved
200 1 dissolved

The preliminary visual experiments showed that the water solubility of the test item is >200 g/l. Therefore the water solubility had to be determined by the “flask method”.

Flask-method

Water solubility of the test item – experiments A to D

Experiment A B C D (Blank)
Amount of test item (mg) 2.5 2.5 2.5 /
Stirring time at 30 °C (h) 24 48 72 72
Aliquot of the flasks μl/10 ml 5/0,01/100/3/10 5/0,01/100/3/10 5/0,01/100/3/10 5/0,01/100/3/10
Measured concentration (μg/ml) 9.5 11.1 11.4 /
Water solubility (g/l) 317 370 380 /

Concentrations of standards for recheck:

Standard Measured concentration
(μg/ml)
Recovery rate
Standard 1 (c = 0.92 μg/ml) 0.91 98.9 %
Standard 2 (c = 2.30 μg/ml) 2.32 100.9 %
Standard 4 (c = 9.20 μg/ml) 9.22 100.2 %

Recovery range from 98,9 % to 100,9 % indicates a sufficient accuracy of the analytical method.

Conclusions:
The water solubility of the test item at a temperature of 20 °C was 380 g/l.
Executive summary:

The water solubility was performed according to OECD guideline 105 using flask method. Based on the study results, the water solubility cs of the test item at a temperature of 20 °C was (without correction for the purity) 380 g/l.
