- Principle of test: RP-HPLC method with charged aerosol detection (CAD)
- Short description of test conditions: hydrogenated tallow/nortallow based IQAC is a mixture of components the determination of the water solubility was
referred to the particular main components. For the determination of the water samples these three substances were analysed. The quantification was based on an external standard calibration derived from the test sample described above. The total water solubility of hydrogenated tallow/nortallow based IQAC was calculated as their sum. Three 5L glass aspirator bottles were rinsed prior to use with a mixture 1:1 methylene chloride : acetone followed by petrolether 30/40. The bottles were dried by a gentle nitrogen flow and rinsed three times with Millipore Type I water. A 4 cm glass-coated stir bar was placed in each bottle. The bottles were placed on magnetic stir plates and filled each with 5L Millipore Type I water. The tests were performed at approximately 22°C. The water was stirred quiescently with no visible vortex (< 90 rpm). After 24 h equilibration time about 250 mg test substance is added to each bottle. Samples were taken after 24 h, 12d , and 14d. Prior sampling the mixing was stopped 1h before. A 150 – 200 ml aliquot was removed from the vessels. The first 100 ml were discarded and the rest was collected in Nalgene bottles. As it was impossible to separate the water phase by centrifugation the solutions were filtrated under pressure with PORAFIL ® PE 0,05 m (part no. 671005047). Beaker, filter, and filtration unit were pre-conditioned with sample solution to avoid sample adsorption. The clear water phases were analysed by HPLC.

Conclusions:
The water solubility of hydrogenated tallow/nortallow based IQAC is considered to be < 0.5 mg/L at 22°C.
Executive summary:

In the present study a HPLC-CAD was performed in order to determine the water solubility of hydrogenated tallow/nortallow based IQAC. Hydrogenated tallow/nortallow based IQAC is composed of three constituents and none of the constituents was detected after 24h, 12 days and 14 days above the detection limits for each constituent. Thus, the water solubility of hydrogenated tallow/nortallow based IQAC is considered to be < 0.5 mg/L at 22°C.
