- - Sampling method: At 0 hour sampling occasion, an aliquot was removed from each of the excess dosing solutions (three aliquots for 0.025 mg/L) and an equivalent volume of tetrahydrofuran (THF) was added (resulting in a x2 dilution). The sample was mixed and an aliquot transferred to an LCMS vial for analysis. At 72 hour sampling occasion, an aliquot was removed from one test vessel from each concentration (three test vessels for 0.025 mg/L) and an equivalent volume of THF was added (resulting in a x2 dilution). - PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A primary stock concentrate of the test item, with a nominal concentration of 10 mg/L, was prepared by weighing a nominal 0.01 g of test substance (actual weight: 0.00998 g) and making up to 1000 mL volume with the culture medium AAP in a volumetric flask. The stock was then stirred for 12 minutes. The resultant stock was a clear and colourless solution and was used to prepare the test solutions. This was achieved by adding the relevant volumes of the primary stock to AAP media and making up to 1000 mL volume in a volumetric flask.

- Observation of abnormalities (for algal test): The microscopic observations, made at the end of the test, showed that compared to the control the algal cells sampled from the nominal 0.007 and 0.013 mg/L test concentrations appeared normal. Algal cells from the nominal 0.025 and 0.05 mg/L appeared small and deformed (very few cells were observed in the 0.05 mg/L concentration). No cells were observed in a sample from the 0.1 mg/L test concentration. - Reported statistics and error estimates:
- The data for cell counts obtained at 24, 48 and 72 hours was entered into electronic data files and statistically analysed using CETIS, where appropriate procedures were applied to test for significant differences (p <0.05) between the control and test concentrations and determine EC50, EC20 and EC10 values and the associated 95% confidence intervals where appropriate. As the measured concentrations at 0 and 72 hours for the nominal 0.007 mg/L were below the limit of quantification (0.006 mg/L), it was not included in the statistical analyses as it could skew the ECX calculations.

The measured concentrations at the start of the study were 0-105% of nominal and at the end were 0-39% of nominal. On this basis the mean measured concentrations were used for the calculation of the effect values.
