Two different predicted data studies have been conducted for target chemical
4-[(4-amino-3-methylphenyl)(4-imino-3-methylcyclohexa-2,5-dien-1-ylidene)methyl]-2-methylaniline hydrochloride (CASno. 3248-91-7) and one experimental study conducted for its read across chemical to determine short term toxicity to aquatic invetebrate endpoint and their results are summarized below.
In first weight of evidence predicted data study the short term toxicity of test chemical 4-[(4-amino-3-methylphenyl)(4-imino-3-methylcyclohexa-2,5-dien-1-ylidene)methyl]-2-methylaniline hydrochloride (CASno. 3248-91-7) was estimated by using OECD QSAR tool box v3.3 with log Kow as primary descriptor and considering the six closest read across substances. When test chemical was exposed for 48 hrs on Daphnia magna EC50 value was estimated to be 508.66 mg/L. Since EC50 value is greater than 100 mg/L it is concluded that this test chemical can be considered to be not classified according to CLP classification criteria.
Another weight of evidence predicted data study done by EPI suite, ECOSAR version 1.1, on the basis of similarity of structure to chemicals for which the aquatic toxicity has been previously measured by structure-activity relationships (SARs) program, the LC 50 value for short term toxicity to aquatic invertebrates was predicted.
On the basis of this program, the LC 50 value for short term toxicity to aquatic invertebrates was predicted to be 385.045 mg/l for test chemical 4-[(4-amino-3-methylphenyl)(4-imino-3-methylcyclohexa-2,5-dien-1-ylidene)methyl]-2-methylaniline hydrochloride (CAS no. 3248-91-7) in 48 hrs.
Based on this value it can be concluded that the substance is considered to be not toxic to aquatic environment and can not be classified in category as per the criteria mentioned in CLP regulation.
Last study was done from authoritative database ( Ecotox. 2017) in this study short term toxicity to Litopenaeus schmitti (Southern white shrimp) was determined for 48 hrs.The study was based on the effects of the read across compound 4 -[(4 -dimethylamino phenyl)-phenylmethylidene]cyclohexa-2,5-dien-1-ylidene]-dimethylazanium chloride (CAS no. 569-64-2) on Litopenaeus schmitti in a static salt water system. Litopenaeus schmitti (Southern white shrimp) was used as a test organism. Based on effect on mortality of the test organism Litopenaeus schmitti, the 48 hr LC50 value was determined to be 77170 mg/L (LC50 value ranges from 60470 to 109120 mg/l).Thus, based on the LC50 value, it can be concluded that the substance 4 -[(4 -dimethylaminophenyl)-phenylmethyli dene]cyclohexa-2,5-dien-1-ylidene]-dimethylazanium chloride can be considered as non-toxic to aquatic organisms and thus can be considered to be not classified as per the CLP classification criteria.
Based on the results of all the studies from different sources mentioned above for target chemical
4-[(4-amino-3-methylphenyl)(4-imino-3-methylcyclohexa-2,5-dien-1-ylidene)methyl]-2-methylaniline hydrochloride (CASno. 3248-91-7) and its read across chemical EC50 value falls in range of 385.045 - 508.66 mg/L and by this range value it is concluded that that test chemical can be considered as not classified according to CLP classification criteria.
