Description of key information

Appearance:

On the basis of physical observation ,the chemical 1-(2-chloroethyl)pyrrolidine hydrochloride was observed to be white coloured Crystalline Powder in appearance.

Melting point:

The melting point determination of 1-(2-chloroethyl)pyrrolidine hydrochloride (CAS No. 7250-67-1) was done following OECD Guideline 102 and according to the metal block capillary method. The test was conducted in replicates and mean melting temperature range was determined to be  in the range of 162.4- 164.2°C at 977.2 hPa.

Boiling point:

The boiling point determination of 1-(2-chloroethyl)pyrrolidine hydrochloride(CAS No. 7250-67-1) was done following OECD Guideline 103 and according to the method of Siwoloboff. The test was conducted in replicates and mean boiling point was determined to be 204°C at 97604 hPa.

Density:

The density of test item 1-(2-chloroethyl)pyrrolidine hydrochloride (CAS No. 7250-67-1)was determined following OECD Guideline 109 and according to the Test methods: Pour density and tapped density of powders” recommended in The International Pharmacopoeia, Fourth Edition, 2014 using bulk density apparartus. The mean pour density and tapped density of test item is was determined to be 0.4001 g/cm³ and 0.5720 g/cm³ at 26.2°C and 978.4 hPa

Particle Size Distribution:

The particle size distribution of test item 1-(2-chloroethyl)pyrrolidine hydrochloride (CAS No. 7250-67-1 ) was determined by granulometric analysis with rotap sieve shaker. The particle size distribution was determined to be in the range of 150 micron to 106 micron.

Vapor Pressure:

Data from various authoritative databases suggests that the estimated vapor pressure of 1-(2-chloroethyl)pyrrolidine hydrochloride at 25 deg C was164 Pa.

Partition Coefficient:

The partition coefficient (n-octanol/water) of test substance was determined as per the HPLC method (OECD Guideline-117). The Log Pow value was determined to be 0.736±0.014 dimensionless at 25°C.

Water Solubility:

The water solubility determination of test item 1-(2-chloroethyl)pyrrolidine hydrochloride (CAS No. 7250-67-1) was done as per the OECD guideline 105 and by spectrophotometric analytical method as per OECD 101. The solubility of 1-(2-chloroethyl)pyrrolidine hydrochloride (CAS:7250-67-1 ) in water was determined to be 500858.301 mg/L at 30°C.

WSKOW v1.42 was used to estimate the water solubility of 1-(2-chloroethyl)pyrrolidine hydrochloride. The estimated water solubility of 1-(2-chloroethyl)pyrrolidine hydrochloride at 25 deg C was 35080 mg/l. The experimental and estimated values indicate that 1-(2-chloroethyl)pyrrolidine hydrochloride is soluble in water.

Surface tension:

Based on the data from ACD labs, surface tension for test item 1-(2-chloroethyl)pyrrolidine hydrochloride was predicted to be 54.9 ± 3.0 dyne/cm.

Flash point:

The flash point determination of test substance 1-(2-chloroethyl)pyrrolidine hydrochloride ( CAS No.7250 -67 -1) was done as per the test guideline OECD 103. The method is closed cup method using Pensky Martin’s Flash Point Apparatus. The flashpoint of the test substance was found to be 61.5°C at 975.6 hPa. Based on criteria mentioned in CLP regulation the chemical has flash point >than 60 degree C hence the substance was considered to be non-flammable.

Autoflammability:

1-(2-chloroethyl)pyrrolidine hydrochloridedid not catch fire on being exposed to air at room temperature of 27°Cpressure of 966 hPa. This indicates that1-(2-chloroethyl)pyrrolidine hydrochlorideis not auto-flammable.

Flammability:

1-(2-chloroethyl)pyrrolidine hydrochloride does not indicate any ignition when the flame of bunsen burner having temperature of approximately 950°C is brought in contact with it. Thus it can be concluded that 1-(2-chloroethyl)pyrrolidine hydrochloride is not flammable at high temperature. However since such high temperatures are not found under normal circumstances of transportation and use, thus 1-(2-chloroethyl)pyrrolidine hydrochloride was considered non-flammable for chemical safety assessment.

Explosivenss:

The study does not need to be conducted because there are no chemical groups present in the molecule which are associated with explosive properties

Oxidising properties:

The study does not need to be conducted because there are no chemical groups present in the molecule which are associated with oxidising properties and hence, the classification procedure does not need to be applied

pH:

The pH determination of test item N-(2-chloroethyl)pyrrolidine hydrochloride (CAS No 7250-67-1) was done following OECD guideline 122. pH of 1% w/v solution was measured with the help of pH meter and it was observed to be 4.34 at 28 °C.

Other supporting study from athoritative databases also indicates that the melting point of chemical 1-(2-chloroethyl)pyrrolidine hydrochloride was determined to be in the range of 173.5 degree C to 174 degree C.

As the data from lab has K1 reliability and the test was performed using OECD guidelines which has the value of melting point temperature as 162.4 degree C to 164.2 degree C thus considered as key study for the chemical safety assessment

The reference substances were Butanone, Acetophenone, Benzophenone, 4-Methoxyphenol, Phenol, Benzene, Biphenyl, Methyl benzoate, Benzyl Benzoate, 1,4-Dichlorobenzene, 1,2,4- Trichlorobenzene, Chlorobenzene, Bromobenzene, 2,3-Dichloroaniline, 4-Acetylpyridine, Acetanilide, 2-Nitrophenol, Nitrobenzene, 2,6-Dichlorobenzonitrile,Diphenylamine, Cinnamyl alcohol, 4-Methylbenzyl alcohol, Benzyl alcohol, Phenoxyacetic acid, Cinnamic Acid, Dodecanoic acid, Toluene, Ethylbenzene, nButylbenzene, Naphthalene, Diphenyl ether, Allyl phenyl ether, DDT having Pow value ranging from 0.3 to 6.5 The Log Pow value was determined to be 0.736±0.014 at 25°C.
