- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)

The test substance was not completely soluble in test medium at the loading rates initially prepared.
Preparation of test solutions started with loading rates ranging from 1.0 to 100 mg/l applying an overnight period of magnetic stirring to reach maximum solubility of the test substance in the test medium. The resulting aqueous mixtures were left to stabilize for 2 to 2¾ hours were after the Water Accommodated Fractions (WAFs) were taken out by siphoning. The lowest test solution for the combined limit/range-finding test was prepared by a 10-fold dilution of the WAF prepared at a loading rate of 1.0 mg/l. All test solutions in the combined limit/range-finding test were clear and colourless besides the highest WAF that was very slightly hazy. In the final test the WAFs prepared at 10 and 18 mg/l were clear and colourless, whereas WAFs prepared at higher loading rates were hazy. - Test organisms (species):

One daphnid was immobilised in the control treatment. Note that the guideline allows up to 10% of immobilisation in the control treatment and therefore, the observed effect was not considered biologically significant.
Note that a floating layer of test substance was observed in all WAFs during the test. However, no test substance was observed on the bodies of the daphnids.
In the three highest groups all daphnids were immobilised. A dose response relationship was observed at the three lowest WAFs. - Results with reference substance (positive control):

The Acute Toxicity Study in Daphnia magna with the test solution was carried out according to OECD 202 Guideline and EU C.2 Method (GLP study).
As the test substance was not completely soluble in test medium at the loading rates initially prepared, the method Water Accommodated Fractions (WAFs) was used. The concentration of test substance in the aquatic samples was calculated using the TOC content of pure test substance.
The 48h-EL50 was between WAFs prepared at loading rates of 10 and 18 mg/L.
