Toxicokinetics :
The physico-chemical parameters as well as the bio-concentration factors can provide reliable indications on the toxico-kinetic behavior of a chemical & whether the chemical shall have a tendency to bio-accumulate. This methodology has been adopted in the absence of any available information related to the toxico-kinetics behaviour of the chemical 5 -Nitrovanillin. Based upon the physico-chemical parameters and bio-concentration factro (BCF) value of 5 -Nitrovanillin that ranges from 3.88 to 4.2, it can be concluded that the chemical shall not have bio-accumulation potential.
Acute Toxicity Effect:
From the predictions done by the QSAR model in the absence of available data for target , the predicted values and supporting values indicate that 5-nitrovanillin is not likely to exhibit acute toxicity by the oral, inhalation and dermal route. Thus, this chemical is not classified as causing acute toxicity by the oral, inhalation and dermal route within the concentration ranges mentioned in the results of the prediction by QSAR.
Skin & eye irritation :
Based upon the results obtained for the irritant effect of 5-nitrovanillin; it is concluded that the chemical has the potential to cause skin and eye irritation. These results also support the majority classification on the CLP inventory and hence it can be conlcuded that the predictions are of reliable quality. Thus, 5 -nitrovanillin is classified as a chemical causing skin and eye irritation
Skin sensitizer :
In the Guinea pig maximization test, as predicted by the QSAR model, the chemical 5-nitrovanillin was found to be non-sensitizing to the skin of guinea pig. Also the supporting studyshows negative results for skin sensitization . Thus, 5 -nitrovanillin is not classifed as a skin sensitizing chemical. This classification is also in agreement with the majority classification in the CLP inventory

DNEL derivation
Sodium 2-amino-4,6-dinitrophenoxide do not exhibit acute toxicity by oral and dermal route of exposure and thus will not be considered in acute toxic category for classification. Sodium 2-amino-4,6-dinitrophenoxide was found to be irritating to skin and eye, thus leading to a conclusion that the target will be irritant to skin as well as eye. Available studies also indicate that the chemical does not exhibit genotoxicity and is not a reproductive toxin within the dose levels mentioned in the end points
In the absence of local effects following short-term or long-term exposure, no dose-response data are available and a quantitative dose descriptor is not derived. DNEL values for local exposure are therefore not calculated.
In the absence of acute systemic toxicity, no dose-response data are available and a quantitative dose descriptor is not derived. DNEL values for acute systemic effects are therefore not calculated.
A standard approach to deriving DNEL values would be to use the repeated dose toxicity dataset and apply assessment factors as described in ECHA guidance documents. The critical endpoint is considered to be the NOAEL of 20000 mg/kg bw/d in oral category.
