Description of key information

A study according to OECD guideline 202 was performed to assess the acute toxicity of the test item to Daphnia magna. Following a preliminary range-finding test, twenty daphnids (4 replicates of 5 animals) were exposed to an aqueous solution of the test item at 0-Hour measured concentrations of 0.34, 0.63, 1.1, 2.1 and 4.2 mg/L for 48 hours at a temperature of 21°C to 22 °C under static test conditions. The 48-Hour EC50 for the test item to Daphnia magna based on 0-Hour measured test concentrations was 3.0 mg/L; 95% confidence limits 2.7 - 3.3 mg/L.

Additional information

Pre-study solubility work conducted indicated that it was not possible to obtain a testable solution of the test item using traditional methods of preparation e.g. ultrasonication. A pre-study media preparation trial indicated that the use of a saturated solution method of preparation was most appropriate for this test item. The test item solution was prepared by stirring an excess (100 mg/L) of test item in test medium using a propeller stirrer at approximately 1500 rpm for 24 hours. After the stirring period any undissolved test item was removed by filtration (0.2 µm Sartorius Sartopore filter, first approximate 2 liters discarded in order to pre-condition the filter) to produce a saturated solution of the test item from which dilutions were made to give the required test concentrations. The number of immobilized Daphnia were recorded after 24 and 48 hours.

Chemical analysis of the test preparations at 0 hours showed measured test concentrations to range from 0.34 to 4.2 mg/L. Analysis of the test preparations at 48 hours showed measured test concentrations to range from 0.32 to 3.8 mg/L (89% to 97% of the 0-Hour measured test concentrations) and hence it was considered appropriate to calculate the results based on the 0-Hour measured test concentrations only.
