There is no study available, in which the short-term toxicity of the target substanceFatty acids, C16-18, stearyl esters (CAS 85536-04-5) to aquatic invertebrates was assessed. Therefore, read-across to the two structurally and chemically closely related source substances Fatty acids, C16-18, 2-hexyldecyl esters (CAS 101227-09-2) and 2-octyldodecyl isooctadecanoate (CAS 93803-87-3) was conducted in accordance with Regulation (EC) No. 1907/2006 Annex XI, 1.5. The source substances are characterized by similar fatty acid chain lengths and alcohol components as the target substance and are therefore considered suitable representatives for the assessment of the short-term toxicity of the target substance to aquatic invertebrates. A detailed justification of the analogue approach is provided in IUCLID section 13.
The available study with the source substance Fatty acids, C16-18, 2-hexyldecyl esters (CAS 101227-09-2) was conducted according to EU method C.2 and in compliance with GLP.
In a static test, Daphnia magna was exposed to two nominal loading rates of 1000 and 3000 mg/L prepared as Water Accommodated Fractions for 48 h. However, the highest loading rate was omitted due to insoluble particles floating on the water surface, which was considered justifiable, since both test concentrations exceed the water solubility of the substance by a factor of > 1000.
After 48 h, no effects on mobility were recorded, resulting in an EL50 (48 h) of > 1000 mg/L (nominal), corresponding to an EL50 (48 h) of > 1.034 mg/L based on measured test item concentrations (GC). Therefore, it was concluded that Fatty acids, C16-18, 2-hexyldecyl esters (CAS 101227-09-2) does not cause short-term toxicity to aquatic invertebrates up to the limit of water solubility.
The available study with the source substance 2-octyldodecyl isooctadecanoate (CAS 93803-87-3) was conducted according to OECD guideline 202 and in compliance with GLP.
In a static limit test, Daphnia magna was exposed to a Water Accommodated Fraction with a nominal loading rate of 86 mg/L test item for 48 h. A chemical analysis of actual test item concentrations was not possible due to the very low water solubility of the test item.
After 48 h, no immobilization was observed, yielding an EL50 (48 h) of > 86.0 mg/L (nominal). Thus, 2-octyldodecyl isooctadecanoate (CAS 93803-87-3) does not cause short-term toxicity to aquatic invertebrates up to the limit of water solubility.
Based on the high degree of structural and chemical similarity between the target and the selected source substances, the target substance is expected to exhibit a similar ecotoxicological profile as the source substances. Therefore, it is concluded that the target substanceFatty acids, C16-18, stearyl esters (CAS 85536-04-5) does not cause short-term toxicity to aquatic invertebrates up to its limit of water solubility (< 0.507 mg/L, 20 °C, OECD 105).
