Description of key information

The acute toxicity of the test substance to the aquatic freshwater invertebrate Daphnia magna was studied under GLP in accordance with OECT TG 202.

The test was conducted under semi-static conditions over a period of 48 hours and test medium was exchanged after 24 hours. Tests were conducted with 100 mL glass beakers covered with glass plates. A volume of 50 mL test medium was added to each beaker and four replicates with five daphnids were tested for each concentration. Water accommodated fractions (WAFs) with the loading rates of 6.25, 12.5, 25, 50 and 100 mg/L were tested, which were prepared by adding defined amounts of substance to reconstituted test water (according to ISO 6341) and stirring for 96 hours at room temperature with subsequent filtration of the dispersions through membrane filters with 0.45 µm pore size. Additionally, a control with test medium without test substance was investigated. The concentrations were measured at the begin and the end of each 24-hour period by using HPLC and geometric means of measured concentrations were calculated for each test period. The mean measured (arithmetric means of each geometric means determined for each test period) test concentrations were 3.3, 5.1 and 8.4 mg/L (the concentrations were not measured for the two highest loading rates). The biological results were based on the loading rates and the mean measured concentrations. The 48-hour EL50 was 11 mg/L based on loading rates and the 48-hour EC50 was 4.7 mg/L based on mean measured concentrations.
