HPLC is performed on analytical columns packed with a commercially available cyanopropyl solid phase containing non-polar and polar moieties.

While passing through the column along with the mobile phase the test substance interacts with the stationary phase. As a result of partitioning between mobile and stationary phases the test substance is retained. The dual composition of the stationary phase having polar and non-polar sites allows for interaction of polar and non-polar groups of a molecule in a similar way as is the case for organic matter in soil or sewage sludge matrices. This enables the relationship between the retention time on the column and the adsorption coefficient on organic matter to be established. According lgKoc and lgk of reference sample and to establish the calibration curve. By this calibration curve, the lgk and lgK0c of test substance were calculated.

Details on results (HPLC method):
Thiourea was used to measure t0. Different concentrations of the reference substances were injected their retention time were recorded. The test solution with a concentration of 1207mg/L was injected and the retention time was determined and recorded. All reference- and test substances were injected and determined two times. Using methanol as the mobile phase, the retention times of the reference samples cannot be linear fit due to the strong eluent strength. According to the sample information, it belongs to no unique and identified molecular structure substance. lt shows two series peaks with the retention times of the peak1 were 2.86minute and 2.86minute respectively and the retention times of the peak2 were 3.69minute and 3.69minute respectively. According to the method, at least one reference substance with a lgKoc value above the expected lgKoc value of test substance should be used.
However, the test substance was very strong retained on the column and its lgKoc was even higher than the reference substance 4,4'-DDT with the highest lgKoc of 5.63 listed in the guideline. The adsorption coefficient (Koc) of the test substance was shown to be very high. Therefore, the lgKoc of the test substance was considered to be greater than 5.63.

Conclusions:
The adsorption Coefficient (Koc) of the test substance was determined by the HPLC method. However, the test substance was very strong retained on the column and its retention time was higher than even that of the reference substance 4,4'-DDT with the highest lgKoc of 5.63. Therefore, the lgKoc of the test substance was considered to be greater than 5.63.
