- Large stocks of the V79 cell line (supplied by Laboratory for Mutagenicity Testing, LMP, Technical University Darmstad, D-64287 Darmstad) were stored in liquid nitrogen in the cell bank of RCC, allowing the repeated use of teh same cell culture batch in experiments. Before freezing each batch was screened for mycoplasm contamination and checked for karyotype stability.
Thawed stock cultures were propagated at 37 °C in 80 cm2 plastic flasks. About 5 * 10^5 cells per flask were seeded into 15 ml of MEM (Minimal Essential Medium) supplemented with 10% fetal calf serum. The cells were subcultured twice weekly. The cell cultures were incubated at 37 °C in a humidified atmosphere with 4.5 % carbon dioxide (95.5% air). - Additional strain / cell type characteristics:

- Test concentrations with justification for top dose:
- A pre-test on cell growth inhibition with 4 and 24 hours treatment was performed in order to determine the toxicity of test item. Test item concentrations between 19.5 and 2500 mg/L were chosen for this evaluation. Since neither too strong toxicity nor precipitation were observed in the pre-test, test item was tested up to 2500 mg/L in the absence and in the presence of S9 mix in both experimental parts.

The substance did not show any potential to induce mutagenic effects both in bacterial and mammalian cells on the basis of results from three different guideline studies, all considered valid.
